Application Notes for anti-PGLYRP1 antibody
Peptide ELISA: Antibody detection limit dilution 1: 4000.
Immunohistochemistry: In paraffin embedded Human Spleen shows staining of select splenocytes. Recommended concentration, 3-6ug/ml.
Western Blot: Approx 30kDa band observed in lysates of cell line Jurkat (calculated MW of 21.7kDa according to NP_005082.1). The observed molecular weight corresponds to earlier findings in literature with different antibodies (Lu et al, J Biol Chem.2006 Mar 3;281(9): 5895-907; PMID: 16354652). Recommended concentration: 0.3-1ug/ml.

Western Blot (WB) of anti-PGLYRP1 antibody

(0.3ug/ml) staining of Jurkat lysate (35ug protein in RIPA buffer). Primary incubation was 1 hour. Detected by chemiluminescence.
Goat Anti-PGLYRP1 Polyclonal Antibody – Cat# MBS421952

Immunohistochemistry (IHC) of anti-PGLYRP1 antibody

(3.8ug/ml) staining of paraffin embedded Human Spleen. Steamed antigen retrieval with citrate buffer pH 6, AP-staining.

Other Notes

Small volumes of anti-PGLYRP1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

UniProt Comments for PGLYRP1

PGLYRP1: Pattern receptor that binds to murein peptidoglycans (PGN) of Gram-positive bacteria. Has bactericidal activity towards Gram-positive bacteria. May kill Gram-positive bacteria by interfering with peptidoglycan biosynthesis. Binds also to Gram- negative bacteria, and has bacteriostatic activity towards Gram- negative bacteria. Plays a role in innate immunity. Belongs to the N-acetylmuramoyl-L-alanine amidase 2 family.

Protein type: Secreted; Receptor, misc.; Apoptosis; Secreted, signal peptide

Chromosomal Location of Human Ortholog: 19q13.2-q13.3

Cellular Component: extracellular region

Molecular Function: N-acetylmuramoyl-L-alanine amidase activity; peptidoglycan binding; peptidoglycan receptor activity; zinc ion binding

Biological Process: defense response to Gram-positive bacterium; detection of bacterium; immune response; innate immune response; negative regulation of inflammatory response; negative regulation of interferon-gamma production; negative regulation of natural killer cell differentiation during immune response; pattern recognition receptor signaling pathway; peptidoglycan catabolic process
