Application Notes for anti-Sirt3 antibody
Peptide ELISA: antibody detection limit dilution 1:64000.
Western blot: Approx 30kDa band observed in Mouse Heart lysates (calculated MW of 28.8kDa according to NP_071878.2). An additional band of unknown identity was also consistently observed at 15kDa. This band was successfully blocked by incubation with the immunizing peptide. Recommended concentration: 1-3µg/ml. Primary incubation 1 hour at room temperature. Preliminary testing was unsuccessful on Rat Kidney for this particular batch.
Immunofluorescence: Strong expression of the protein seen in the cytoplasm of NIH3T3 cells. Recommended concentration: 10µg/ml.
Flow Cytometry: Flow cytometric analysis of NIH3T3 cells. Recommended concentration: 10ug/ml.

Western Blot (WB) of anti-Sirt3 antibody

(0.3ug/ml) staining of Mouse Heart lysate (35ug protein in RIPA buffer). Primary incubation was 1 hour. Detected by chemiluminescence.

Immunofluorescence (IF) of anti-Sirt3 antibody

Immunofluorescence analysis of paraformaldehyde fixed NIH3T3 cells, permeabilized with 0.15% Triton. Primary incubation 1hr (10ug/ml) followed by Alexa Fluor 488 secondary antibody (2ug/ml), showing cytoplasmic staining. The nuclear stain is DAPI (blue). Negative control: Unimmunized goat IgG (10ug/ml) followed by Alexa Fluor 488 secondary antibody (2ug/ml).

Flow Cytometry (FC) of anti-Sirt3 antibody

Flow cytometric analysis of paraformaldehyde fixed NIH3T3 cells (blue line), permeabilized with 0.5% Triton, showing a low level of staining. Primary incubation 1hr (10ug/ml) followed by Alexa Fluor 488 secondary antibody (1ug/ml). IgG control: Unimmunized goat IgG (black line) followed by Alexa Fluor 488 secondary antibody.

Other Notes

Small volumes of anti-Sirt3 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

UniProt Comments for Sirt3

SIRT3: a soluble mitochondrial NAD-dependent protein deacetylase. Can deacetylate and thereby activate a central metabolic regulator in the mitochondrial matrix, glutamate dehydrogenase. Mitochondrial proteins are hyperacetylated in SIRT3-deficient mice, but not in SIRT4 or SIRT5 deficient mice. SIRT3-deficient mice have no obvious metabolic phenotype, and the process of adaptive thermogenesis is normal. Can deacetylate and activate isocitrate dehydrogenase 2, an enzyme that participates in regulation of the citric acid cycle and the regeneration of antioxidants. Inhibited by nicotinamide, but not by trichostatin A. Its unprocessed form is enzymatically inactive. It is processed by mitochondrial processing peptidase (MPP) in the mitochondrial matrix to give an enzymatically active 28 kDa product.

Protein type: Deacetylase; Mitochondrial; EC 3.5.1.-

Cellular Component: cytoplasm; membrane; mitochondrial inner membrane; mitochondrial matrix; mitochondrion

Molecular Function: (3,5-dichlorophenylurea)acetate amidohydrolase activity; enzyme binding; hydrolase activity; hydrolase activity, acting on carbon-nitrogen (but not peptide) bonds, in linear amides; indoleacetamide hydrolase activity; iprodione amidohydrolase activity; metal ion binding; N-acetylgalactosamine-6-phosphate deacetylase activity; N2-acetyl-L-lysine deacetylase activity; NAD binding; NAD-dependent histone deacetylase activity (H3-K14 specific); O-succinylbenzoate synthase activity; protein binding; protein N-terminal asparagine amidohydrolase activity; UDP-3-O-[3-hydroxymyristoyl] N-acetylglucosamine deacetylase activity; zinc ion binding

Biological Process: aerobic respiration; positive regulation of insulin secretion; protein amino acid deacetylation
