Application Notes for anti-POU2AF1 antibody
Peptide ELISA: Antibody detection limit dilution 1: 8000.
Immunohistochemistry: In paraffin embedded Human Spleen shows nuclear staining in the germinal center with stronger nucleoli staining in some of the B cells. Recommended concentration, 3-5ug/ml.
Western Blot: In transfected HEK293 transiently expressing full-length Human POU2AF1 (myc and DYKDDDDK tagged), a band of approx.38kDa was observed. No bands were observed in mock-transfected HEK293 and the same band was observed using anti-DYKDDDDK tag antibody. Recommended concentration, 1-2ug/ml.

Western Blot (WB) of anti-POU2AF1 antibody

HEK293 lysate (10ug protein in RIPA buffer) overexpressing Human POU2AF1 with DYKDDDDK tag probed with in Lane A and probed with anti- DYKDDDDK Tag (1/3000) in lane C. Mock-transfected HEK293 probed with in Lane B. Primary incubations were for 1 hour. Detected by chemiluminescence.

POU2AF1: Transcriptional coactivator that specifically associates with either OCT1 or OCT2. It boosts the OCT1 mediated promoter activity and to a lesser extent, that of OCT2. It has no intrinsic DNA-binding activity. It recognizes the POU domains of OCT1 and OCT2. It is essential for the response of B-cells to antigens and required for the formation of germinal centers. A chromosomal aberration involving POU2AF1/OBF1 may be a cause of a form of B-cell leukemia. Translocation t(3;11)(q27;q23) with BCL6. Belongs to the POU2AF1 family.
