Immunoprecipitation (IP) of anti-SF3B2 antibody

SF3B2 was immunoprecipitated from 1mg of THP1 cells membrane fraction, blotted with M25215 of 10 ug. Western blot was performed from the immunoprecipitate using M25215 at 1/1000 dilution. Anti-Mouse-IgG(HRP), specific to the non-reduced form of IgG, was used as secondary antibody at 1/10000 dilution. Lane 1:THP1 cells membrane fraction. Lane2:IP product of THP1 cells membrane fraction.

Blocking and dilution buffer and concentration:5% milk/TBST.

Flow Cytometry (FC/FACS) of anti-SF3B2 antibody

Flow cytometric analysis of 2% paraformaldehyde-fixed THP1 (Human acute monocytic leukemia cell line) cells labeling SF3B2 with M25215 at 1/200 dilution (red) compared with a mouse monoclonal IgG isotype control (black) and an unlabelled control (cells without incubation with primary antibody, blue). Goat anti-mouse IgG (FITC) at 1/300 dilution was used as the secondary antibody.

UniProt Comments for SF3B2

Involved in pre-mRNA splicing as a component of the splicing factor SF3B complex (PubMed:27720643). SF3B complex is required for 'A' complex assembly formed by the stable binding of U2 snRNP to the branchpoint sequence (BPS) in pre-mRNA. Sequence independent binding of SF3A/SF3B complex upstream of the branch site is essential, it may anchor U2 snRNP to the pre-mRNA (PubMed:12234937). May also be involved in the assembly of the 'E' complex (PubMed:10882114). Belongs also to the minor U12-dependent spliceosome, which is involved in the splicing of rare class of nuclear pre-mRNA intron (PubMed:15146077).
