PABP2/ PABPN1 (PABP2/PABPN1) Antibody – Mouse Monoclonal

For Research Use Only. Not for use in diagnostic procedures. Catalog #: MBS435140

This gene encodes an abundant nuclear protein that binds with high affinity to nascent poly(A) tails. The protein is required for progressive and efficient polymerization of poly(A) tails at the 3' ends of eukaryotic transcripts and controls the size of the… More details

Western Blot (WB) of anti-PABP2/PABPN1 antibody

All lanes : PABP2 Mouse mAb at 1/2000 dilution Lane 1 : THP1 cells membrane fraction Lane 2 : Jurkat cells membrane fraction Lysates/proteins at 20 ug per lane. Secondary Goat Anti-Mouse IgG-HRP, 5% milk conjugated at 1/10000 dilution Predicted band size : 33 KDa Observed band size : 50 KDa Blocking/Dilution buffer : 1x TBST.
Mouse Anti-PABP2/PABPN1 Monoclonal Antibody – Cat# MBS435140

Immunoprecipitation (IP) of anti-PABP2/PABPN1 antibody

PABP2 was immunoprecipitated from 1mg of THP1 cells membrane fraction, blotted with M25211 of 10 ug. Western blot was performed from the immunoprecipitate using M25211 at 1/1000 dilution. Anti-Mouse-IgG(HRP), specific to the non-reduced form of IgG, was used as secondary antibody at 1/10000 dilution. Lane 1: THP1 cells membrane fraction. Lane2: IP product of THP1 cells membrane fraction.

Blocking and dilution buffer and concentration:5% milk/TBST.

Flow Cytometry (FC/FACS) of anti-PABP2/PABPN1 antibody

Flow cytometric analysis of 2% paraformaldehyde-fixed THP1 (Human acute monocytic leukemia cell line) cells labeling PABP2 with M25211 at 1/200 dilution (red) compared with a mouse monoclonal IgG isotype control (black) and an unlabelled control (cells without incubation with primary antibody, blue). Goat anti-mouse IgG (FITC) at 1/300 dilution was used as the secondary antibody.

Other Notes

Small volumes of anti-PABP2/PABPN1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

UniProt Comments for PABP2/PABPN1

Involved in the 3'-end formation of mRNA precursors (pre-mRNA) by the addition of a poly(A) tail of 200-250 nt to the upstream cleavage product (). Stimulates poly(A) polymerase (PAPOLA) conferring processivity on the poly(A) tail elongation reaction and controls also the poly(A) tail length (). Increases the affinity of poly(A) polymerase for RNA (). Is also present at various stages of mRNA metabolism including nucleocytoplasmic trafficking and nonsense-mediated decay (NMD) of mRNA. Cooperates with SKIP to synergistically activate E-box-mediated transcription through MYOD1 and may regulate the expression of muscle-specific genes (PubMed:11371506). Binds to poly(A) and to poly(G) with high affinity (). May protect the poly(A) tail from degradation (). Subunit of the trimeric poly(A) tail exosome targeting (PAXT) complex, a complex that directs a subset of long and polyadenylated poly(A) RNAs for exosomal degradation. The RNA exosome is fundamental for the degradation of RNA in eukaryotic nuclei. Substrate targeting is facilitated by its cofactor MTREX, which links to RNA-binding protein adapters (PubMed:27871484).
