200ug/ml of Ab purified from Bioreactor Concentrate by Protein A/G. Prepared in 10mM PBS with 0.05% BSA & 0.05% azide. Also available WITHOUT BSA & azide at 1.0mg/ml.
Specificity
Recognizes a dimer of two proteins of 70kDa and ~80kDa, identified as two subunits of Ku. MAb KU729 recognizes a conformational epitope of p70/p80 dimer, which is destroyed during Western blotting. The p70/p80 dimer is important for function of a 460kDa DNA-dependent protein kinase. Ku protein plays a role in cell signaling, proliferation, DNA repair, replication, transcriptional activation, and apoptosis.

Immunogen
Nuclear extract of human HL-60 cells
Quantity and Buffer
0.5 ml at 200ug/ml with BSA and azide
Applications Tested/Suitable for anti-XRCC6 (p70) antibody
Immunofluorescence (IF), Immunoprecipitation (IP), Immunocytochemistry (ICC), Immunohistology (IH)
Application Notes for anti-XRCC6 (p70) antibody
Immunofluorescence (0.5-1.0ug/ml); Immunoprecipitation (0.5-1ug/500ug protein lysate); Immunocytochemistry (Acetone-fixed cells) (0.25-0.5ug/ml for 30 minutes at RT) Immunohistology (Frozen & Formalin-fixed) (0.5-1.0ug/ml for 30 minutes at RT) (Staining of formalin-fixed tissues requires boiling tissue sections in 10mM Citrate Buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 minutes) Optimal dilution for a specific application should be determined.

Other Notes

Small volumes of anti-XRCC6 (p70) antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for XRCC6 (p70)

The p70/p80 autoantigen is a nuclear complex consisting of two subunits with molecular masses of approximately 70 and 80 kDa. The complex functions as a single-stranded DNA-dependent ATP-dependent helicase. The complex may be involved in the repair of nonhomologous DNA ends such as that required for double-strand break repair, transposition, and V(D)J recombination. High levels of autoantibodies to p70 and p80 have been found in some patients with systemic lupus erythematosus. [provided by RefSeq, Jul 2008]
