Human, Rat, Mouse
Form/Format
Rabbit polyclonal, affinity-purified antibody is supplied in 100 ul phosphate-buffered saline, 50% glycerol, 1 mg/ml BSA, and 0.05% sodium azide.
Purity/Purification
Affinity Purified
Specificity
The antibody detects a 95kDa* protein corresponding to the apparent molecular mass of Sema3A on SDS-PAGE immunoblots of human recombinant Sema3A and neonatal rat brain lysates.
Preparation and Storage
Store at -20°C. Stable for 1 year.
Chromosome Location
Chromosome: 7; NC_000007.13 (83587659..83824217, complement). Location: 7p12.1
OMIM
603961
3D Structure
ModBase 3D Structure for Q14563
Immunogen
Semaphorin 3A synthetic peptide (coupled to carrier protein) corresponding to amino acids in the central region of human Sema3A. The sequence used is highly conserved in rat and mouse Sema3A, and has low homology to other semaphorin family members.
Specificity Note
All molecular weights (MW) are confirmed by comparison to MW standards and to western blot mobilities of known proteins with similar MW.
"Native" western blot utilizes non-reducing sample buffer (no mercaptoethanol or SDS), normal SDS-PAGE gel electrophoresis, and no methanol in transfer buffers.
Applications Tested/Suitable for anti-SEMA3A antibody
ELISA, Western Blot (WB), Immunohistochemistry (IHC), Immunocytochemistry (ICC)
Application Notes for anti-SEMA3A antibody
ELISA: 1:2000
Western Blot (WB): 1:1000
Immunohistochemistry (IHC): 1:300
Immunocytochemistry (ICC): 1:100
End user should determine optimal dilution for their particular applications and experiments. Western blot membranes were incubated with diluted antibody in 5% non-fat milk, Tris buffer, 0.04% Tween20 for 1 hour at room temperature.

Testing Data of anti-SEMA3A antibody

Western blots of neonatal rat brain (lanes 1, 3 & 5) and human recombinant Sema3A/Fc chimera (95/125 kDa) (lanes 2, 4 & 6). Blots were probed with anti-Sema3A ( MBS472059 ) (lanes 1 & 2), anti-Sema3A (MBS472057) (lanes 3 & 4) and anti-Sema3A ( MBS472058 ) (lane 5 & 6). The antibodies recognize both the 95 kDa and 125 kDa forms of the recombinant Sema3A.

Test Data of anti-SEMA3A antibody

Formalin fixed, citric acid treated parafin sections of adult Rat cerebellum. Sections were probed with anti-Sema3A (MBS472057) then anti-RabbitHRP before detection using DAB. (Images provided by Carl Hobbs and Dr. Pat Doherty at Wolfson Centre for Age-Related Diseases, King's College London).

NCBI Summary for SEMA3A

This gene is a member of the semaphorin family and encodes a protein with an Ig-like C2-type (immunoglobulin-like) domain, a PSI domain and a Sema domain. This secreted protein can function as either a chemorepulsive agent, inhibiting axonal outgrowth, or as a chemoattractive agent, stimulating the growth of apical dendrites. In both cases, the protein is vital for normal neuronal pattern development. Increased expression of this protein is associated with schizophrenia and is seen in a variety of human tumor cell lines. Also, aberrant release of this protein is associated with the progression of Alzheimer's disease. [provided by RefSeq]
