Product Overview

Online Datasheet in PDF
Product Name
Dynamin (Ser774) (DNM1), Polyclonal Antibody
Full Product Name
Anti-Phospho-Ser774 Dynamin
Product Gene Name
Clonality
Polyclonal
Host
Sheep
Species Reactivity
Rat
Form/Format
100 ul in 10 mM HEPES (pH 7.5), 150 mM NaCl, 100 ug BSA per ml and 50% glycerol. Adequate amount of material to conduct 10-mini Western Blots.
Purity/Purification
Affinity Purified (Prepared from sheep serum by affinity purification via sequential chromatography on phospho- and dephosphopeptide affinity columns.)
Specificity
Specific for the ~95k dynamin protein phosphorylated at Ser774. Labels the purified protein phosphorylated in vitro by cdk5 but not by PKC. Does not cross react with other purified substrates of cdk5 (e.g. amphiphysin and synapsin). Immunolabeling is blocked by lambda-phosphatase treatment.
Preparation and Storage
For long term storage -20 degree C is recommended. Stable at -20 degree C for at least 1 year.
Molecular Weight
95
Chromosome Location
Chromosome: 3; NC_005102.3 (16686951..16730913, complement). Location: 3p11
3D Structure
ModBase 3D Structure for P21575
Antigen
Phosphopeptide corresponding to amino acid residues surrounding the phospho-Ser774 of rat dynamin.
Immunogen Information
Synthetic phospho-peptide corresponding to amino acid residues surrounding Ser774 conjugated to KLH
Immunogen Species
Rat
Reactivity Assumed Based on 100% Sequence Homology
Bovine, canine, chicken, human, mouse and non-human primates
Species Reactivity Note
The antibody has been directly tested for reactivity in Western blots with rat tissue. It is anticipated that the antibody will react with bovine, canine, chicken, human, mouse and non-human primates based on the fact that these species have 100% homology with the amino acid sequence used as antigen.
Biological Significance
Dynamin is a member of a group of nerve terminal proteins called dephosphins that regulate synaptic vesicle endocytosis (Cousin et al., 2001; Graham et al., 2002; Tsuboi et al., 2002). Cyclin dependent protein kinase 5 phosphorylates dynamin at Ser774 and Ser778 that are the phosphorylation sites on dynamin phosphorylated in vivo (Tan et al., 2003). Phosphorylation of Ser774 by GSK3 has recently been shown to control activity-dependent bulk endocytosis of synaptic vesicles (Clayton et al., 2010)
Applications Tested/Suitable for anti-DNM1 antibody
Western Blot (WB), Immunofluorescence (IF), Immunohistochemistry (IHC)
Application Notes for anti-DNM1 antibody
Quality Control: Western blots performed on each lot.
WB: 1:1000

Other Notes

Small volumes of anti-DNM1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
