CPB2 Antibody – Sheep Polyclonal

For Research Use Only. Not for use in diagnostic procedures. Catalog #: MBS512148

TAFI (Thrombin Activatable Fibrinolysis Inhibitor), also referred to as plasma procarboxypeptidase-B, procarboxypeptidase-U and R, circulates in plasma as a zymogen with a mass of 58,000 daltons (1-6). Proteolytic activation of TAFI yields an N-terminally… More details

Sheep
Form/Format
IgG-HRP conjugate as a clear, slightly red-brown liquid.
Purity/Purification
Peroxidase conjugated IgG.
Vial containing ml of whole IgG conjugated to horseradish peroxidase (HRP) through carbohydrate groups. Total protein is 0.2 mg.
Concentration
IgG-HRP concentration is mg/ml, determined by absorbance using an extinction coefficient (E1%280) of 14. (lot specific)
Specificity
Prior to conjugation, this antibody was specific for TAFI as demonstrated by immunoelectrophoresis and ELISA.

Other Notes

Small volumes of anti-Cpb2 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

UniProt Comments for Cpb2

Function: Cleaves C-terminal arginine or lysine residues from biologically active peptides such as kinins or anaphylatoxins in the circulation thereby regulating their activities. Down-regulates fibrinolysis by removing C-terminal lysine residues from fibrin that has already been partially degraded by plasmin.

Catalytic activity: Release of C-terminal Arg and Lys from a polypeptide.

Cofactor: Binds 1 zinc ion per subunit

Enzyme regulation: TAFI/CPB2 is unique among carboxypeptidases in that it spontaneously inactivates with a short half-life, a property that is crucial for its role in controlling blood clot lysis. The zymogen is stabilized by interactions with the activation peptide. Release of the activation peptide increases a dynamic flap mobility and in time this leads to conformational changes that disrupt the catalytic site and expose a cryptic thrombin-cleavage site present at Arg-323

Subcellular location: Secreted Ref.2.

Tissue specificity: Plasma; synthesized in the liver. Ref.2

Sequence similarities: Belongs to the peptidase M14 family.
