Human, Bovine
Form/Format
Lyophilized from 20mM Potassium Phosphate, pH 7.2, with 150mM Sodium Chloride, 10mg/ml BSA as a stabilizer (Immunoglobulin and Protease free), and 0.01% Sodium Azide added as a preservative. Reconstitute with 0.1ml of deionized water (or equivalent).
Purity/Purification
Collagen Type IV antibody (biotin) was purified by affinity chromatography.
Concentration
100ug, lyophilized (lot specific)
Specificity
Bovine/Human collagen type IV
Preparation and Storage
Ships ambient or refrigerated. Upon receipt store at 4°C while lyophilized. After reconstituted, store at 4°C short term, or aliquot and store at -20°C long term. Avoid repeat freeze-thaws.
Tag/Conjugate
Biotin
Immunogen
Collagen type IV from human and bovine placenta
Species Reactivity Note
This antibody reacts with most mammalian Type IV collagens and has negligible cross reactivity with Type I, II, III, IV collagens. Non specific cross reaction with other human serum proteins or non-collagen extracellular matrix proteins is negligible.
Bioactivity
Some class-specific anti-collagens may be specific for three-dimensional epitopes which may result in diminished reactivity with denatured collagen or formalin-fixed, paraffin embedded tissues
Biohazard Information
This product contains sodium azide as preservative. Although the amount of sodium azide is very small appropriate care must be taken when handling this product.
Applications Tested/Suitable for anti-COL4 antibody
ELISA, Immunohistochemistry (IHC), Western Blot (WB)
Application Notes for anti-COL4 antibody
ELISA: 1:20,000-1:100,000
IHC: 1:1,000-1:5,000
WB: 1:2,000-1:10,000

Immunohistochemistry of anti-COL4 antibody

Immunohistochemical staining using Collagen Type IV antibody
Anti collagen IV antibody (1:400, 45 min RT) showed strong staining in FFPE sections of human kidney (Left) with strong red staining observed in glomeruli and liver (Right) with strong staining in sinusoids. Staining for both tissues was consistent with a basement membrane distribution. Slides were steamed in 0. 01 M sodium citrate buffer, pH 6. 0 at 99-100 deg C - 20 minutes for antigen retrieval.

Other Notes

Small volumes of anti-COL4 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI GeneID

NCBI Summary for COL4

This gene encodes one of the six subunits of type IV collagen, the major structural component of basement membranes. Mutations in this gene are associated with X-linked Alport syndrome, also known as hereditary nephritis. Like the other members of the type IV collagen gene family, this gene is organized in a head-to-head conformation with another type IV collagen gene so that each gene pair shares a common promoter. Alternatively spliced transcript variants have been identified for this gene. [provided by RefSeq, Aug 2010]

UniProt Comments for COL4

COL4A5: Type IV collagen is the major structural component of glomerular basement membranes (GBM), forming a 'chicken-wire' meshwork together with laminins, proteoglycans and entactin/nidogen. Defects in COL4A5 are the cause of Alport syndrome X- linked (APSX). APSX is characterized by progressive glomerulonephritis, renal failure, sensorineural deafness, specific eye abnormalities (lenticonous and macular flecks), and glomerular basement membrane defects. The disorder shows considerable heterogeneity in that families differ in the age of end-stage renal disease and the occurrence of deafness. Deletions covering the N-terminal regions of COL4A5 and COL4A6, which are localized in a head-to-head manner, are found in the chromosome Xq22.3 centromeric deletion syndrome. This results in a phenotype with features of diffuse leiomyomatosis and Alport syndrome (DL-ATS). Belongs to the type IV collagen family. 2 isoforms of the human protein are produced by alternative splicing.
