Interleukin-1 Receptor Antagonist (IL1RN) Human Active Protein

For Research Use Only. Not for use in diagnostic procedures. Catalog #: MBS546089

Interleukin-1 receptor antagonist (IL-1ra) is a member of the IL-1 family. Endogenous IL-1ra is produced in numerous animal disease models as well as in human autoimmune and chronic inflammatory diseases. It binds to IL-1 receptors in competition with IL-1,… More details

Product Overview

Product Name
Interleukin-1 receptor antagonist (IL1RN), Active Protein
Full Product Name
Recombinant Human Interleukin-1 receptor antagonist (rHu IL-1ra)
Product Gene Name
Product Synonym Names
Interleukin-1 receptor antagonist; human IL-1RA; Human Interleukin-1 receptor antagonist
Host
E Coli
Form/Format
Lyophilized from a 0.2mm filtered concentrated (1mg/ml) solution in PBS, pH 7.4.
Physical Appearance: Sterile Filtered White lyophilized (freeze-dried) powder.
Purity/Purification
>95% by SDS-PAGE and HPLC analyses.
Preparation and Storage
This lyophilized preparation is stable at 2-8 degree C, but should be kept at -20 degree C for long term storage, preferably desiccated. Upon reconstitution, the preparation is stable for up to one week at 2-8 degree C. For maximal stability, apportion the reconstituted preparation into working aliquots and store at -20 degree C to -70 degree C. Avoid repeated freeze/thaw cycles.
Molecular Weight
20,055 Da
Chromosome Location
Chromosome: 2; NC_000002.11 (113868693..113891593). Location: 2q14.2
OMIM
147679
3D Structure
ModBase 3D Structure for P18510
Active Protein Note
Protein activity can be an ambiguous term. Please refer to the data sheet or inquire with Technical Support to determine the definition of activity for this product.
Molecular Weight Information
Approximately 16.5 kDa, a single non-glycosylated polypeptide chain containing 156 amino acids.
Biological Activity
Fully biologically active when compared to standard. The ED50 as determined by the dose-dependant inhibition of IL-1 stimulation of D10S cells was found to be 0.5 ng/ml, corresponding to a Specific Activity of 2.0 x 106 IU/mg.
Endotoxin Level
Less than 1EU/mg of rHuIL-1ra as determined by LAL method.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Reconstitute in sterile distilled water or aqueous buffer containing 0.1% BSA to a concentration of 0.1-1.0 mg/mL. Stock solutions should be apportioned into working aliquots and stored at <-20 degree C. Further dilutions should be made in appropriate buffered solutions.

Other Notes

Small volumes of IL1RN active protein vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for IL1RN

The protein encoded by this gene is a member of the interleukin 1 cytokine family. This protein inhibits the activities of interleukin 1, alpha (IL1A) and interleukin 1, beta (IL1B), and modulates a variety of interleukin 1 related immune and inflammatory responses. This gene and five other closely related cytokine genes form a gene cluster spanning approximately 400 kb on chromosome 2. A polymorphism of this gene is reported to be associated with increased risk of osteoporotic fractures and gastric cancer. Four alternatively spliced transcript variants encoding distinct isoforms have been reported. [provided by RefSeq, Jul 2008]

UniProt Comments for IL1RN

IL1RN: Inhibits the activity of interleukin-1 by binding to receptor IL1R1 and preventing its association with the coreceptor IL1RAP for signaling. Has no interleukin-1 like activity. Binds functional interleukin-1 receptor IL1R1 with greater affinity than decoy receptor IL1R2; however, the physiological relevance of the latter association is unsure. The intracellular form of IL1RN is predominantly expressed in epithelial cells. 4 isoforms of the human protein are produced by alternative splicing.
