MIP-1 Beta (CCL4) (MIP-1 beta) Human Recombinant Protein

For Research Use Only. Not for use in diagnostic procedures. Catalog #: MBS553044

Macrophage Inflammatory Protein-1 is a factor produced by macrophages that causes local inflammatory responses, and induces superoxide production by neutrophils. Two peptides are responsible for this activity. They have been termed MIP-1-alpha, and MIP-… More details

Form/Format
Recombinant MIP-1 beta was lyophilized from a 0.2 um filtered PBS solution.
Purity/Purification
>95%, as determined by SDS-PAGE and HPLC
Preparation and Storage
The lyophilized protein is stable for at least 2 years from date of receipt at -20 degree C.
Upon reconstitution, this cytokine can be stored in working aliquots at 2 degree - 8 degree C for one month, or at -20 degree C for six months, with a carrier protein without detectable loss of activity.

Avoid repeated freeze/thaw cycles.

Host Note
Optimized DNA sequence encoding Human MIP-1 beta chain was expressed in Escherichia Coli.
Endotoxin
Endotoxin content was assayed using a LAL gel clot method.
Endotoxin level was found to be less than 0.1 ng/ug (1EU/ug).
Reconstitution
A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers
Molecular Weight Note
Native human MIP-1 beta is generated by the proteolytic removal of the signal peptide and propeptide. This molecule has a calculated molecular mass of approximately 8 kDa.

Recombinant MIP-1 beta is a disulfide-linked homodimeric protein consisting of 70 amino acid residue subunits, and migrates as an approximately 8 kDa protein under non-reducing and reducing conditions in SDS-PAGE.

Other Notes

Small volumes of MIP-1 beta recombinant protein vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for MIP-1 beta

This gene is one of several cytokine genes that are clustered on the q-arm of chromosome 17. Cytokines are a family of secreted proteins that function in inflammatory and immunoregulatory processes. The protein encoded by this family member is similar to the chemokine (C-C motif) ligand 4 product, which inhibits HIV entry by binding to the cellular receptor CCR5. The copy number of this gene varies among individuals, where most individuals have one to five copies. This gene copy contains a non-consensus splice acceptor site at the 3' terminal exon found in other highly similar gene copies, and it thus uses other alternative splice sites for the 3' terminal exon, resulting in multiple transcript variants. UniProt Comments for MIP-1 beta

CCL4: Monokine with inflammatory and chemokinetic properties. Binds to CCR5. One of the major HIV-suppressive factors produced by CD8+ T-cells. Recombinant MIP-1-beta induces a dose-dependent inhibition of different strains of HIV-1, HIV-2, and simian immunodeficiency virus (SIV). The processed form MIP-1-beta(3-69) retains the abilities to induce down-modulation of surface expression of the chemokine receptor CCR5 and to inhibit the CCR5- mediated entry of HIV-1 in T-cells. MIP-1-beta(3-69) is also a ligand for CCR1 and CCR2 isoform B. Belongs to the intercrine beta (chemokine CC) family.
