Host
E Coli (Host protein expression may be lot-specific and can be subject to change during production, including bulk orders)
Form/Format
Recombinant PEDF was lyophilized from a 0.2 um filtered 20mM PB,150mM NaCl solution pH 7.5.
Purity/Purification
>95%, as determined by SDS-PAGE and HPLC
Preparation and Storage
The lyophilized protein is stable for at least 2 years from date of receipt at -20 degree C.
Upon reconstitution, this cytokine can be stored in working aliquots at 2 degree - 8 degree C for one month, or at -20 degree C for six months, with a carrier protein without detectable loss of activity.

Avoid repeated freeze/thaw cycles.

Host Note
Optimized DNA sequence encoding Human PEDF mature chain was expressed in Escherichia Coli.
Endotoxin
Endotoxin content was assayed using a LAL gel clot method.
Endotoxin level was found to be less than 0.1 ng/ug (1EU/ug).
Reconstitution
A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.
Molecular Weight Note
Native human PEDF, generated by the proteolytic removal of the signal peptide and propeptide, the molecule has a calculated molecular mass of approximately 45kDa.

Recombinant PEDF is a disulfide-linked monomer protein consisting of 440 amino acid residue subunits, and migrates as an approximately 45kDa protein under non-reducing and reducing conditions in SDS-PAGE.

Other Notes

Small volumes of PEDF recombinant protein vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for PEDF

The protein encoded by this gene is a member of the serpin family, although it does not display the serine protease inhibitory activity shown by many of the other serpin family members. The encoded protein is secreted and strongly inhibits angiogenesis. In addition, this protein is a neurotrophic factor involved in neuronal differentiation in retinoblastoma cells.[provided by RefSeq, Mar 2011]

UniProt Comments for PEDF

PEDF: a secreted neurotrophic protein that induces extensive neuronal differentiation in retinoblastoma cells. Potent inhibitor of angiogenesis. As it does not undergo the S (stressed) to R (relaxed) conformational transition characteristic of active serpins, it exhibits no serine protease inhibitory activity. The N-terminal (AA 44-121) exhibits neurite outgrowth-inducing activity. The C-terminal exposed loop (AA 382-418) is essential for serpin activity. Extracellular phosphorylation enhances antiangiogenic activity.
