Platelet Derived Growth Factor-Aa (PDGFAA) Human Recombinant Protein

For Research Use Only. Not for use in diagnostic procedures. Catalog #: MBS553119

PDGF is synthesized mainly by megakaryocytes. It is stored in the alpha granules of platelets from which it is released after cell activation of platelets. Platelets synthesize a mixture of the three possible isoforms (BB,AB,AA) while fibroblasts stimulated… More details

Host Note
Optimized DNA sequence encoding Human PDGF-AA mature chain was expressed in Escherichia Coli.
Endotoxin
Endotoxin content was assayed using a LAL gel clot method.
Endotoxin level was found to be less than 0.1 ng/ug (1EU/ug).
Reconstitution
A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.
Molecular Weight Note
Human PDGF-AA, is generated by the proteolytic removal of the signal peptide
and propeptide. The molecule has a calculated molecular mass of approximately 28 kDa.

Recombinant PDGF-AA is a disulfide-linked homodimeric protein consisting of two 126 amino acid residue subunits, migrates as an approximately 28 kDa protein under non-reducing and as 14kDa under reducing conditions in SDS-PAGE.

Other Notes

Small volumes of PDGFAA recombinant protein vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for PDGFAA

The protein encoded by this gene is a member of the platelet-derived growth factor family. The four members of this family are mitogenic factors for cells of mesenchymal origin and are characterized by a motif of eight cysteines. This gene product can exist either as a homodimer or as a heterodimer with the platelet-derived growth factor beta polypeptide, where the dimers are connected by disulfide bonds. Studies using knockout mice have shown cellular defects in oligodendrocytes, alveolar smooth muscle cells, and Leydig cells in the testis; knockout mice die either as embryos or shortly after birth. Two splice variants have been identified for this gene. [provided by RefSeq, Jul 2008]

UniProt Comments for PDGFAA

PDGFA: Growth factor that plays an essential role in the regulation of embryonic development, cell proliferation, cell migration, survival and chemotaxis. Potent mitogen for cells of mesenchymal origin. Required for normal lung alveolar septum formation during embryogenesis, normal development of the gastrointestinal tract, normal development of Leydig cells and spermatogenesis. Required for normal oligodendrocyte development and normal myelination in the spinal cord and cerebellum. Plays an important role in wound healing. Signaling is modulated by the formation of heterodimers with PDGFB. Belongs to the PDGF/VEGF growth factor family. 2 isoforms of the human protein are produced by alternative splicing.
