Form/Format
Lyophilized; PBS, pH 7.2
Purity/Purification
>95%, as determined by SDS-PAGE and HPLC.
Preparation and Storage
The lyophilized protein is stable for at least 2 years from date of receipt at -20 degree C. Upon reconstitution, this product can be stored in working aliquots at 2 degree C - 8 degree C for one month, or at -20 degree C for six months, with a carrier protein without detectable loss of activity. Avoid repeated freeze/thaw cycles.
Molecular Weight
42,905 Da
Active Protein Note
Protein activity can be an ambiguous term. Please refer to the data sheet or inquire with Technical Support to determine the definition of activity for this product.
Species
Mouse
Source Note
Optimized DNA sequence encoding Mouse WNT3A mature chain including a C-terminal 6xHis tag was expressed in CHO. Recombinant Mouse Protein Wnt-3a encodes amino acid residues 19-352.
Biological Activity
Determined by its ability to induced alkaline phosphatase production by CCL-226 cells. The expected ED50 for this effect is 0.4-0.6 ng/ml.
Endotoxin
Endotoxin content was assayed using a LAL gel clot method. Endotoxin level was found to be less than 0.1 ng/ug(1EU/ug).
Reconstitution
A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.
Applications Tested/Suitable for WNT3A active protein
SDS-PAGE
Application Notes for WNT3A active protein
Recombinant WNT3A migrates as an approximately 37 kDa protein due to glycosylation under reducing conditions in SDS-PAGE.

Other Notes

Small volumes of WNT3A active protein vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for WNT3A

The WNT gene family consists of structurally related genes which encode secreted signaling proteins. These proteins have been implicated in oncogenesis and in several developmental processes, including regulation of cell fate and patterning during embryogenesis. This gene is a member of the WNT gene family. It encodes a protein which shows 96% amino acid identity to mouse Wnt3A protein, and 84% to human WNT3 protein, another WNT gene product. This gene is clustered with WNT14 gene, another family member, in chromosome 1q42 region. [provided by RefSeq, Jul 2008]

UniProt Comments for WNT3A

Ligand for members of the frizzled family of seven transmembrane receptors (Probable). Functions in the canonical Wnt signaling pathway that results in activation of transcription factors of the TCF/LEF family (PubMed:20093360, PubMed:21244856, PubMed:24841207, PubMed:26902720). Required for normal embryonic mesoderm development and formation of caudal somites. Required for normal morphogenesis of the developing neural tube (). Mediates self-renewal of the stem cells at the bottom on intestinal crypts (in vitro) (PubMed:26902720).
