Mouse, Rat
Form/Format
Lyophilized 100 ul with 0.09% sodium azide as a preservative.
Preparation and Storage
Maintain at 2-8°C for 3 months or at -20°C for longer periods. Stable for 1 year. Avoid repeated freeze-thaw cycles.
Immunogen Sequence
Sulfated CCK-8 (26-33) coupled to bovine thyroglobulin (BTg) with glutaraldehyde.
Reconstitution
Do not reconstitute until ready to use since the product is most stable when lyophilized. The product does not need to be cooled during shipping. For long-term storage, store lyophilized antibody until ready to use at -15° C or lower. Reconstitute with 100 uL of distilled or deionized water.

Application Notes for anti-CCK-8 antibody
Immunofluroescence: 1:100-1:200 (Fluorescein)
1:500-1:1,000 (Streptavidin/HRP).

Tissue Preparation:
10 um cryostats
Fixative: 4% paraformaldehyde in 0.1 M phosphate buffer, pH 7.4; 500 mL over ~20-30 min.
Post Fixation: 1.5 hr. at 4°C in 4% paraformaldehyde in 0.1 M phosphate buffer, pH 7.4.

Immunohistochemistry:
The antibody has siginificant indirect immunofluorescent staining at a 1/100-1/200 dilution and significant Biotin-Streptavidin/HRP immunostaining at a 1/500-1/1,000 dilution in rat hypothalamus and spinal cord. The specificity of the antiserum was examined by soluble pre-adsorption with the peptides in question at a final concentration of 10-6M CCK-8 immunolabeling was completely abolished by pre-adsorption with CCK-8, gastrin 17 and gastrin 34. Pre-adsorption with the following peptides resulted in no reduction of immunostaining: alphaACGRP, BETACGRP, neurotensin, somatostatin, substance P, leucine enkephalin, methionine enkephalin, VIP, neuropeptide Y, gastric inhibitory polypeptide, bombesin, glucagon, peptide YY, and FMRF amide.
Optimal dilution should be determined by investigator.

Other Notes

Small volumes of anti-CCK-8 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
