Immunocytochemistry: 1:20-1:50 on acetone or formaldehyde fixed cytospin preparations.
Immunohistochemistry (Frozen sections): 1:20 on acetone fixed frozen sections followed by incubation with IgG rabbit anti-mouse and a monoclonal mouse APAAP complex. Alternatively, after incubation with the primary antibody (1:20) and washing, slides can be incubated with IgG (Biotin) rabbit anti-mouse and Streptavidin (HRP).

Immunofluorescence (IF) of anti-ABCB4 antibody

Immunolocalization of MRP3 in 2008 cells and MDCKII monolayers by confocal laser scanning microscopy. MRP3 is detected by indirect immunofluorescence (green signal) with MBS600967. Nucleic acids were detected by counterstaining with propidium iodide (red signal). (A) Parental 2008 cells; (B) 2008/MRP3–8 cells; (C) parental MDCKII cells; (D) MDCKII/MRP3–17 cells; (E) vertical X/Z section of the monolayer shown in D. The arrow in D indicates the position where the X/Z section was made. (Bar = 20um.)

Immunostaining (IS) of anti-ABCB4 antibody

Immunohistochemical staining of human liver. (A and B) Immunohistochemical detection of MRP3 in the lateral membranes of cholangiocytes (indicated by arrows) and (baso) lateral membranes of hepatocytes surrounding the portal tracts by using MBS600967. B is an enlargement of A. (C) Immunohistochemical detection of cytokeratin 7 in bile-duct epithelium cells. (D) Immunostaining with M2III-6 against MRP2, showing canalicular staining of the hepatocytes through out the entire liver lobule.

Other Notes

Small volumes of anti-ABCB4 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for ABCB4

The membrane-associated protein encoded by this gene is a member of the superfamily of ATP-binding cassette (ABC) transporters. ABC proteins transport various molecules across extra- and intra-cellular membranes. ABC genes are divided into seven distinct subfamilies (ABC1, MDR/TAP, MRP, ALD, OABP, GCN20, White). This protein is a member of the MDR/TAP subfamily. Members of the MDR/TAP subfamily are involved in multidrug resistance as well as antigen presentation. This gene encodes a full transporter and member of the p-glycoprotein family of membrane proteins with phosphatidylcholine as its substrate. The function of this protein has not yet been determined; however, it may involve transport of phospholipids from liver hepatocytes into bile. Alternative splicing of this gene results in several products of undetermined function. [provided by RefSeq]

UniProt Comments for ABCB4

ABCB4: a transporter that mediates ATP-dependent export of organic anions and drugs from the cytoplasm. Hydrolyzes ATP with low efficiency. Human ABCB4 is not capable of conferring drug resistance. Mediates the translocation of phosphatidylcholine across the canalicular membrane of the hepatocyte. Interacts with HAX1. Defects are the cause of gallstones, and progressive familial intrahepatic cholestasis 3, an autosomal recessive liver disorder that progresses to cirrhosis and liver failure before adulthood. Two alternatively spliced human isoforms have been described.
