Application Notes for anti-JAK3 antibody
Dilution: Immunoprecipitation: 5-10ug immunoprecipitated JAK3 from 2mg of RIPA lysate from CTLL cells, previously this antibody has immunoprecipitated JAK3 from a TF-1 RIPA cell lysate.
Immunoprecipitation Kinase Assay: 5-10ug of this antibody immunoprecipitates JAK3, which can be autophosphorylated in an in vitro kinase reaction. TF1 cell lysate was immunoprecipitated with J0902 (10ug). The immune complex was incubated in kinase buffer containing [y-32 P]ATP and subsequently resolved by electrophoresis, followed by autoradiography.

Immunoprecipitation (IP) of anti-JAK3 antibody

Immunoprecipitation Kinase Assay: Representative blot from a previous lot. TF1 cell lysate was immunoprecipitated with MBS603010 (10ug). Arrow indicates JAK3 (~125kD)

Other Notes

Small volumes of anti-JAK3 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for JAK3

The protein encoded by this gene is a member of the Janus kinase (JAK) family of tyrosine kinases involved in cytokine receptor-mediated intracellular signal transduction. It is predominantly expressed in immune cells and transduces a signal in response to its activation via tyrosine phosphorylation by interleukin receptors. Mutations in this gene are associated with autosomal SCID (severe combined immunodeficiency disease). [provided by RefSeq]

UniProt Comments for JAK3

JAK3: a non-receptor tyrosine-kinase of the Jak family expressed predominantly in immune cells. Mediates signal transduction via the common gamma-chain of cytokine receptors, including IL-2, -4, -7, -9, and -15. Interacts with members of the STAT (signal transduction and activators of transcription) family. Interacts with STAM2 and SHB. Contains two protein kinase domains. Mutations that abrogate JAK3 function cause an autosomal SCID (severe combined immunodeficiency disease). Inhibitor: R017s for organ transplants. Three differentially spliced isoforms have been described.
