Application Notes for anti-RANGAP1 antibody
Peptide ELISA: antibody detection limit dilution >1:32,000. Western Blot: Approx 70kD band observed in Human Brain and 3T3 lysates (predicted MW of 68kD according to NP_002874). Recommended for use at 0.1-0.5ug/ml., Staining (0.2ug/ml) of 3T3 lysate (RIPA buffer, 35ug total protein per lane). Primary incubated for 1 hour. Detected by western blot using chemiluminescence. Optimal dilutions to be determined by the researcher.

Other Notes

Small volumes of anti-RANGAP1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for RANGAP1

This gene encodes a protein that associates with the nuclear pore complex and participates in the regulation of nuclear transport. The encoded protein interacts with Ras-related nuclear protein 1 (RAN) and regulates guanosine triphosphate (GTP)-binding and exchange. Alternative splicing results in multiple transcript variants. UniProt Comments for RANGAP1

RANGAP1: a cytoplasmic GTPase activator for the Ras-related regulatory protein Ran, converting it to the putatively inactive GDP-bound state. Antagonist of RCC1, a regulator molecule that keeps RAN in the active, GTP-bound state. RanGAP1 and RCC1 control RAN-dependent transport between the nucleus and cytoplasm. RanGAP1 is a key regulator of the RAN GTP/GDP cycle. Homodimerizes. Forms a tight complex in association with RANBP2 and the ubiquitin-conjugating enzyme E2 (UBC9).

Protein type: GAPs, Ras; GAPs

Chromosomal Location of Human Ortholog: 22q13

Cellular Component: kinetochore; spindle pole; nuclear membrane; perinuclear region of cytoplasm; cytoplasm; nuclear pore; cytosol

Molecular Function: protein binding

Biological Process: negative regulation of protein export from nucleus; mitotic cell cycle; signal transduction
