Application Notes for anti-TNR antibody
Western Blot: This antibody can be used at 0.1-0.2ug/ml with the appropriate secondary reagents to detect human Tenascin R. The detection limit for rhTenascin R is approximately 2 ng/lane and 20 ng/lane under non-reducing and reducing conditions, respectively., Immunohistochemistry: This antibody will detect human Tenascin R in cells and tissues. The working dilution is 2-5ug/ml., Direct ELISA: This antibody can be used at 0.5-1.0ug/ml with the appropriate secondary reagents to detect human Tenascin R. The detection limit for rhTenascin R is approximately 0.3 ng/well., Optimal dilutions should be determined by each laboratory for each application.

Immunohistochemistry (IHC) of anti-TNR antibody

Tenascin R in Human Brainstem. Tenascin R was detected in immersion fixed paraffin-embedded sections of human brainstem (medulla) using 1.7ug/ml MBS614768 overnight at 4°C. Tissue was stained with the Anti-Goat HRP-DAB Cell & Tissue Staining Kit (brown) and counterstained with hematoxylin (blue).

Other Notes

Small volumes of anti-TNR antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

Subunit structure: Forms homodimers and homotrimers. Interacts with CNTN1, NFASC and CSPG5. Ref.1 Ref.2 Ref.3 Ref.4 Ref.5

Subcellular location: Secreted › extracellular space › extracellular matrix. Note: May be attach to the cell surface of neural cells.

Tissue specificity: Brain specific. Ref.1

Developmental stage: Expression weakly detectable at E6 embryo, reaches a maximum at E16 and declines in the adult. Ref.1

Domain: The N-terminus cysteine-rich segment may mediate the formation of oligomers. The fibronectin type-III 2-3 mediate the binding to contactin 1. The fibronectin type-III 9 mediates the cell attachment. The fibronectin type-III 2-5 mediate NFASC binding. The fibrinogen C-terminal domain mediates interaction with CSPG5. Ref.5
