Application Notes for anti-EHMT2 antibody
Suitable for use in Immunoblot.
Dilution: Western Blot: 0.2-2ug/ml detected G9a in HeLa nuclear extract cell lysate. HeLa nuclear extract was resolved by electrophoresis, transferred to nitrocellulose and probed with anti-G9a (0.5ug/ml). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system.

Other Notes

Small volumes of anti-EHMT2 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for EHMT2

A cluster of genes, BAT1-BAT5, has been localized in the vicinity of the genes for TNF alpha and TNF beta. This gene is found near this cluster; it was mapped near the gene for C2 within a 120-kb region that included a HSP70 gene pair. These genes are all within the human major histocompatibility complex class III region. This gene was thought to be two different genes, NG36 and G9a, adjacent to each other but a recent publication shows that there is only a single gene. The protein encoded by this gene is thought to be involved in intracellular protein-protein interaction. There are three alternatively spliced transcript variants of this gene but only two are fully described. [provided by RefSeq]

UniProt Comments for EHMT2

EHMT2 iso4: Histone methyltransferase. Preferentially methylates 'Lys-9' of histone H3 and 'Lys-27' of histone H3 (in vitro). H3 'Lys-9' methylation represents a specific tag for epigenetic transcriptional repression by recruiting HP1 proteins to methylated histones. Probably targeted to histone H3 by different DNA-binding proteins like E2F6, MGA, MAX and/or DP1. Also methylates histone H1. Belongs to the histone-lysine methyltransferase family. Suvar3-9 subfamily. Part of the E2F6.com-1 complex in G0 phase composed of E2F6, MGA, MAX, TFDP1, CBX3, BAT8, EUHMTASE1, RING1, RNF2, MBLR, L3MBTL2 and YAF2. Interacts with GFI1B, WIZ and EHMT1. 3 isoforms of the human protein are produced by alternative splicing.

Protein type: EC 2.1.1.43; Amino Acid Metabolism - lysine degradation; Methyltransferase, protein lysine; Methyltransferase

Chromosomal Location of Human Ortholog: 6p21.31

Cellular Component: nucleoplasm; intermediate filament cytoskeleton; intracellular membrane-bound organelle; nuclear chromatin; nucleus

Molecular Function: protein binding; zinc ion binding; p53 binding; protein-lysine N-methyltransferase activity; histone lysine N-methyltransferase activity (H3-K9 specific); histone-lysine N-methyltransferase activity; histone lysine N-methyltransferase activity (H3-K27 specific)

Biological Process: histone methylation; establishment and/or maintenance of chromatin architecture; peptidyl-lysine di-methylation; DNA methylation; negative regulation of transcription from RNA polymerase II promoter; regulation of DNA replication; histone H3-K9 methylation
