Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Application Notes for anti-H3 antibody
Suitable for use in Western Blot, Immunocytochemistry and Chromatin Immunoprecipitation.
Dilution: Western Blot: 0.05ug/ml detected Histone H3 on 10ug of HeLa acid extract, sodium butyrate treated HeLa cells and recombinant Histone H3. Proteins were visualized using an IgG (HRP) goat anti-rabbit and a chemilumnescence detection system.
Immunocytochemistry: 1:500 detected acetylated Histone H3 in NIH/3T3 and A431 cells.
Chromatin Immunoprecipitation: 5ug using HeLa extracted chromain

Chemiluminescence of anti-H3 antibody

HeLa acid extract (lane1), Sodium butyrate treated HeLa cells (lane 2) and recombinant Histone H3 (Lane3) lysates were probed with MBS617139 (0.05ug/ml). Proteins were visualized using a anti-rabbit IgG conjugated to HRP and a chemiluminescence detection system. Arrow indicates Histone H3 (~17kD).

Immunofluorescence (IF) of anti-H3 antibody

Confocal fluorescent analysis of A431 and NIH/3T3 cells using MBS617139 (Red). Actin filaments have been labeled with AlexaFluor488- Phalloidin (Green). Nucleus is stained with DAPI (Blue). This antibody positively stains the nucleus.
