Applications Tested/Suitable for anti-NRP1 antibody
Flow Cytometry (FC/FACS)
Application Notes for anti-NRP1 antibody
Suitable for use in Flow Cytometry.
Dilution: Flow Cytometry: 10ul labels 1-2.5x10e5 cells. Validated on bEND.3 cells and PC-12 cells.
Flow Cytometry Protocol: 1. Cells may be Fc-blocked with 1ug of mouse IgG/10e5 cells for 15 minutes at RT. Do not wash excess blocking IgG from this reaction.
2. After blocking, 10ul of N1276-91C was added to 1-2.5x10e5 cells and incubated for 30 minutes at RT.
3. Unbound antibody was removed by washing the cells
twice in Flow Cytometry Staining Buffer . Note that whole blood requires a RBC lysis step at this point using Flow Cytometry Mouse Lyse Buffer.
4. The cells were resuspended in Flow Cytometry Staining
Buffer for analysis by Flow Cytometry. As a control for this
analysis, cells in a separate tube should be treated with
CFS-labeled goat IgG antibody.

Other Notes

Small volumes of anti-NRP1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for NRP1

NRP1 is a membrane-bound coreceptor to a tyrosine kinase receptor for both vascular endothelial growth factor (VEGF; MIM 192240) and semaphorin (see SEMA3A; MIM 603961) family members. NRP1 plays versatile roles in angiogenesis, axon guidance, cell survival, migration, and invasion.[supplied by OMIM]

UniProt Comments for NRP1

NRP1: The membrane-bound isoform 1 is a receptor involved in the development of the cardiovascular system, in angiogenesis, in the formation of certain neuronal circuits and in organogenesis outside the nervous system. It mediates the chemorepulsant activity of semaphorins. It binds to semaphorin 3A, The PLGF-2 isoform of PGF, The VEGF-165 isoform of VEGF and VEGF-B. Coexpression with KDR results in increased VEGF-165 binding to KDR as well as increased chemotaxis. It may regulate VEGF-induced angiogenesis. Homodimer, and heterodimer with NRP2. Interacts with FER. Binds PLXNB1. The expression of isoforms 1 and 2 does not seem to overlap. Isoform 1 is expressed by the blood vessels of different tissues. In the developing embryo it is found predominantly in the nervous system. In adult tissues, it is highly expressed in heart and placenta; moderately in lung, liver, skeletal muscle, kidney and pancreas; and low in adult brain. Isoform 2 is found in liver hepatocytes, kidney distal and proximal tubules. Belongs to the neuropilin family. 2 isoforms of the human protein are produced by alternative splicing.
