Application Notes for anti-BNIP3 antibody
Suitable for use in ELISA, Western Blot, Immunofluorescence, and Immunohistochemistry.
Dilution: ELISA: 1:1,000
Western Blot: 1:100-1:500
Immunohistochemistry: 1:50-1:100
Immunofluorescence: 1:50-1:100

Immunofluorescence (IF) of anti-BNIP3 antibody

Fluorescent confocal image of HepG2 cells stained with BNIP3 (BH3 Domain Specific) antibody. HepG2 cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.2%, 30 min). Cells were then incubated with BNIP3 (BH3 Domain Specific) primary antibody (1:500, 2 h at room temperature). For secondary antibody, Alexa Fluor® 488 conjugated donkey anti-rabbit antibody (green) was used (1:1000, 1h). Nuclei were counterstained with Hoechst 33342 (blue) (10 μg/ml, 5 min). BNIP3 immunoreactivity is localized to the cytoplasm of HepG2 cells.
Rabbit Anti-BNIP3 Polyclonal Antibody – Cat# MBS625815

Western Blot (WB) of anti-BNIP3 antibody

The anti-NIP3 BH3 domain Pab is used in Western blot to detect NIP3 BH3 in Ramos cell lysate (lane 1) and in mouse brain tissue lysate (lane 2).

Immunofluorescence (IF) of anti-BNIP3 antibody

Freshly isolated mouse hepatocytes plated on coverslips (2 x105 cells/22-mm glass coverslip) were cultured under normoxic conditions for 6 hr. The cells were then fixed in 2% paraformaldehyde in PBS for 1 hr, and processed for confocal immunofluorescence (red: F-actin, blue: ATP-synthase, green: BNIP3). Fluorescence labeling of BNIP3 accomplished with anti-BNIP3 antibody Cat # AP1321a. Data courtesy of Ruben Zamora, University of Pittsburgh.
Rabbit Anti-BNIP3 Polyclonal Antibody, Immunofluorescence (IF) – Cat# MBS625815

Immunohistochemistry (IHC) of anti-BNIP3 antibody

Formalin-fixed and paraffin-embedded human cancer tissue reacted with the primary antibody, which was peroxidase-conjugated to the secondary antibody, followed by DAB staining. This data demonstrates the use of this antibody for immunohistochemistry; clinical relevance has not been evaluated. BC = breast carcinoma; HC = hepatocarcinoma.

NCBI Summary for BNIP3

This gene is a member of the BCL2/adenovirus E1B 19 kd-interacting protein (BNIP) family. It interacts with the E1B 19 kDa protein, which protects cells from virally-induced cell death. The encoded protein also interacts with E1B 19 kDa-like sequences of BCL2, another apoptotic protector. This protein contains a BH3 domain and a transmembrane domain, which have been associated with pro-apoptotic function. The dimeric mitochondrial protein encoded by this gene is known to induce apoptosis, even in the presence of BCL2. [provided by RefSeq]

UniProt Comments for BNIP3

BNIP3: Apoptosis-inducing protein that, which can overcome BCL2 suppression. May play a role in repartitioning calcium between the two major intracellular calcium stores in association with BCL2. Homodimer. Binds to BCL2. Interacts with BNIP3L and ACAA2. Also can interact with adenovirus E1B 19 kDa protein or Epstein-Barr virus BHRF1. Belongs to the NIP3 family.
