Dilution: Peptide ELISA Titer: 1:16,000
Western Blot: 0.3-1ug/ml ~100kD band observed in Human Lung lysates (calculated MW of 100kD according to isoform 1, NP_001074.2). In transfected HEK293 transiently expressing PDE5A isoform 2 a band of approx. 95kD is observed. This band is not observed in the non-transfected HEK293 (calculated MW of 94.8kD according to isoform 2, NP_236914.2).

PDE5A: a cGMP-binding, cGMP-specific phosphodiesterase, a member of the cyclic nucleotide phosphodiesterase family. This phosphodiesterase specifically hydrolyzes cGMP to 5'-GMP. It is involved in the regulation of intracellular concentrations of cyclic nucleotides and is important for smooth muscle relaxation in the cardiovascular system. Phosphorylation is regulated by binding of cGMP to the two allosteric sites. Sildenafil (Viagra) is a highly selective and potent inhibitor of PDE5A. Alternative splicing results in four transcript variants encoding distinct isoforms.
