MDM2 Antibody – Rabbit Polyclonal

For Research Use Only. Not for use in diagnostic procedures. Catalog #: MBS626571

MDM2 is a nuclear phosphoprotein with an apparent molecular mass of 90kD that forms a complex with the p53 tumor suppressor protein. Human MDM2 was identified as a homologous product of the 'murine double minute 2' gene (mdm2). The MDM2 gene enhances the… More details

Host
Rabbit
Species Reactivity
Mouse
Form/Format
Supplied as a liquid in PBS, pH 7.2, 0.1% sodium azide. No stabilizing proteins added.
Purity/Purification
Affinity Purified
Purified by immunoaffinity chromatography.
Specificity
Recognizes mouse MDM 2 showing a band at ~102kD corresponding to phosphorylated MDM2. Cellular Localization: Nuclear and cytoplasmic. Expressed predominantly in the nucleoplasm. Interaction with ARF (P14) results in the localization of both proteins to the nucleus. The nucleolar localization signals in both ARF and MDM2 may be necessary to allow efficient nucleolar localization of both proteins.
Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degree C. Aliquots are stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Other Notes

Small volumes of anti-MDM2 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for MDM2

This gene is a target gene of the transcription factor tumor protein p53. The encoded protein is a nuclear phosphoprotein that binds and inhibits transactivation by tumor protein p53, as part of an autoregulatory negative feedback loop. Overexpression of this gene can result in excessive inactivation of tumor protein p53, diminishing its tumor suppressor function. This protein has E3 ubiquitin ligase activity, which targets tumor protein p53 for proteasomal degradation. This protein also affects the cell cycle, apoptosis, and tumorigenesis through interactions with other proteins, including retinoblastoma 1 and ribosomal protein L5. More than 40 different alternatively spliced transcript variants have been isolated from both tumor and normal tissues. [provided by RefSeq]

UniProt Comments for MDM2

MDM2: a ubiquitin ligase for p53, plays a central role in regulation of the stability of p53 via Akt-mediated MDM2 phosphorylation. Phosphorylation of MDM2 increases its interaction with p300, providing a platform to allow the assembly of the protein complex necessary for MDM2-mediated ubiquitination and degradation of p53. Phosphorylation of MDM2 also blocks its binding to p19ARF, increasing the degradation of p53. Facilitates the nuclear export of p53 and targets it for proteasome-mediated proteolysis. Eight alternatively spliced isoforms have been reported.
