Application Notes for anti-PRKCA antibody
Suitable for use in ELISA, Western Blot and Immunoprecipitation.
Dilution: Western Blot: A 1:1,000 dilution of the lot detected PKC alpha phosphorylated on residues Ser657 and Tyr658 in immunoprecipitated lysate from neonatal rat brain. Similar results resulted in using other lysates such as SKN-SH, HeLa, human endothelial, rabbit spleen
fibroblast, and rat pituitary cells.

Immunoprecipitation (IP) of anti-PRKCA antibody

Immunoprecipitation: 5-10 μL have been shown by an independent lab to work in IP. For IP, this antibody detects isoforms alpha and beta, but no other PKC isoforms. Immunoblot Analysis Immunoprecipitated lysate of neonatal rat brain were resolved by electrophoresis, transferred to PVDF, and probed with anti-phospho-PKC (Ser657/Tyr658) for 2 hours at room temperature. Lane 2 was pre-treated with alkaline phosphatase. Proteins were visualized via HRP and chemiluminescent detection. Arrow indicates phospho-PKC (Ser657/Tyr658).Lower bands are IgG from IP.

Other Notes

Small volumes of anti-PRKCA antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for PRKCA

Protein kinase C (PKC) is a family of serine- and threonine-specific protein kinases that can be activated by calcium and the second messenger diacylglycerol. PKC family members phosphorylate a wide variety of protein targets and are known to be involved in diverse cellular signaling pathways. PKC family members also serve as major receptors for phorbol esters, a class of tumor promoters. Each member of the PKC family has a specific expression profile and is believed to play a distinct role in cells. The protein encoded by this gene is one of the PKC family members. This kinase has been reported to play roles in many different cellular processes, such as cell adhesion, cell transformation, cell cycle checkpoint, and cell volume control. Knockout studies in mice suggest that this kinase may be a fundamental regulator of cardiac contractility and Ca(2+) handling in myocytes. [provided by RefSeq]

UniProt Comments for PRKCA

PKCA: an AGC kinase of the PKC family. A classical PKC downstream of many mitogenic and receptors. Classical PKCs are calcium-dependent enzymes that are activated by phosphatidylserine, diacylglycerol and phorbol esters. Contains a pseudo-substrate autoinhibitory domain that binds to the catalytic domain preventing its activation in the absence of cofactors or activators.
