Product Categories/Family for anti-SAE1 antibody

Applications Tested/Suitable for anti-SAE1 antibody
ELISA (EL/EIA), Western Blot (WB)
Application Notes for anti-SAE1 antibody
Suitable for use in ELISA and Western Blot.
Dilution: ELISA: 1:100,000-1:125,000
Western Blot: 1:750-1:1,000

Western Blot (WB) of anti-SAE1 antibody

Western blot using Immunochemicals’ Rabbit-anti-SAE1 pS185 antibody shows detection of phosphorylated SAE1. Left lane (-) contains 20 _g human HeLa whole cell protein. Right lane (+) contains 20 _g human HeLa whole cell protein from cells pre-treated with phosphatase inhibitor cocktail to prevent dephos- phorylation of the target. Proteins were separated on a 10% SDS-PAGE and transferred onto nitrocellulose. After blocking with 5% milk-TBST 1 hr at room temperature, the membrane was probed with the primary antibody diluted to 1:1,000 at room temperature for 3 hr followed by washes and reaction with HRP-conjugated secondary and ECL imaging.

Other Notes

Small volumes of anti-SAE1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for SAE1

Posttranslational modification of proteins by the addition of the small protein SUMO (see SUMO1; MIM 601912), or sumoylation, regulates protein structure and intracellular localization. SAE1 and UBA2 (MIM 613295) form a heterodimer that functions as a SUMO-activating enzyme for the sumoylation of proteins (Okuma et al., 1999 [PubMed 9920803]).[supplied by OMIM]

UniProt Comments for SAE1

UBLE1A: The heterodimer acts as a E1 ligase for SUMO1, SUMO2, SUMO3, and probably SUMO4. It mediates ATP-dependent activation of SUMO proteins followed by formation of a thioester bond between a SUMO protein and a conserved active site cysteine residue on UBA2/SAE2. Belongs to the ubiquitin-activating E1 family.
