Western Blot (WB) of anti-CAMK2B antibody

Western Blot analysis of mouse brain lysate|Lane 1: C1036-78 |Lane 2: MBS632090
Mouse Anti-CAMK2B Monoclonal Antibody – Cat# MBS632090

Western Blot (WB) of anti-CAMK2B antibody

Western Blot analysis was performed on whole cell extracts (30ug lysate) of SH-SY5Y (Lane 1), U-87MG (Lane 2) and PC12 (Lane 3). The blots were probed withMBS632090 (1:1000) and detected by chemiluminescence using IgG (HRP) Gtxi-Mo. A 70kD band corresponding to CaM Kinase II beta was observed only in PC12. As it is a Rat immunogen, it did not react with human cell lines.

Flow Cytometry (FC/FACS) of anti-CAMK2B antibody

Flow Cytometry analysis of U-87 MG cells. Cells were fixed with 70% ethanol for 10 min, permeabilized with 0.25% Triton™ X-100 for 20 min, and blocked with 5% BSA for 30 min at RT. Cells were labeled withMBS632090 (red histogram) at a dilution of 1:20 or a matched isotype control (pink histogram) in 2.5% BSA. After incubation at RT for 2 hours, the cells were labeled with Alexa Fluor® 488 Rabbit Anti-Mouse Secondary Antibody for 30 min at RT. The purple histogram represents unstained control cells and the green histogram represents no-primary-antibody control and the pink histogram represents isotype control.

Other Notes

Small volumes of anti-CAMK2B antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for CAMK2B

The product of this gene belongs to the serine/threonine protein kinase family and to the Ca(2+)/calmodulin-dependent protein kinase subfamily. Calcium signaling is crucial for several aspects of plasticity at glutamatergic synapses. In mammalian cells, the enzyme is composed of four different chains: alpha, beta, gamma, and delta. The product of this gene is a beta chain. It is possible that distinct isoforms of this chain have different cellular localizations and interact differently with calmodulin. Eight transcript variants encoding eight distinct isoforms have been identified for this gene. [provided by RefSeq, Jul 2008]

UniProt Comments for CAMK2B

CAMK2B: a widely expressed protein kinase of the calcium/calmodulin-dependent protein kinase (CAMK) group. Functions autonomously after Ca2+/calmodulin-binding and autophosphorylation. Involved in dendritic spine and synapse formation, neuronal plasticity and regulation of sarcoplasmic reticulum Ca2+ transport in skeletal muscle. In neurons, plays an essential structural role in the reorganization of the actin cytoskeleton during plasticity by binding and bundling actin filaments in a kinase-independent manner. This structural function is required for correct targeting of CaMK2A, which acts downstream of NMDAR to promote dendritic spine and synapse formation and maintain synaptic plasticity which enables long-term potentiation (LTP) and hippocampus-dependent learning. In developing hippocampal neurons, promotes arborization of the dendritic tree and in mature neurons, promotes dendritic remodeling. Participates in the modulation of skeletal muscle function in response to exercise. In slow-twitch muscles, is involved in regulation of sarcoplasmic reticulum (SR) Ca2+ transport and in fast-twitch muscle participates in the control of Ca2+ release from the SR through phosphorylation of triadin, a ryanodine receptor-coupling factor, and phospholamban, an endogenous inhibitor of SERCA2. The holoenzyme is composed of 4 different chains: alpha (CAMK2A), beta (CAMK2B), gamma (CAMK2G), and delta (CAMK2D). The different isoforms assemble into homo- or heteromultimeric holoenzymes composed of 12 subunits with two hexameric rings stacked one on top of the other. Interacts with SYNGAP1 and CAMK2N2. Interacts with MPDZ. Expressed in adult and fetal brain. Expression is slightly lower in fetal brain. Expressed in skeletal muscle and induced during exercise. Eight isoforms of the human protein are produced by alternative splicing
