P27kip1 Antibody – Mouse Monoclonal

For Research Use Only. Not for use in diagnostic procedures. Catalog #: MBS632206

The cyclin-dependent kinase inhibitor p27KIP1 is a potent inhibitor of both Cyclin E-Cdk2 and Cyclin A-Cdk2, two cdk complexes involved in the regulation of G1/S transition. p27 is phosphorylated on threonine 187 by cyclin E/Cdk2, and overexpression of this… More details

Product Name
p27kip1, Monoclonal Antibody
Full Product Name
p27kip1, phosphorylated (Thr187)
Product Synonym Names
Anti -p27kip1, phosphorylated (Thr187)
Clonality
Monoclonal
Isotype
IgG1,k
Clone Number
6A153
Host
Mouse
Species Reactivity
Hamster, Mouse, Rat
Form/Format
Supplied as a liquid in PBS, pH 7.4, containing 0.1% sodium azide
Purity/Purification
Affinity Purified
Purified by Protein A chromatography.
Specificity
Specific for p27 when phosphorylated at Thr187. On Western blots, it identifies the target band at ~27kD. It does not react with unphosphorylated p27. Specificity has been confirmed with Thr187-mutant p27 recombinant protein. Reactivity has been confirmed with p27-transfected 293 cells. Based on sequence homology, this antibody may also react with mouse, rat and hamster.
Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Synthetic peptide encompassing the phosphorylated Thr187 residue of human p27
Applications Tested/Suitable for anti-p27kip1 antibody
ELISA (EL/EIA), Western Blot (WB)
Application Notes for anti-p27kip1 antibody
Suitable for use in ELISA and Western Blot.
Dilution: ELISA: 0.1-1ug/ml
Western Blot: 1-3ug/ml

Other Notes

Small volumes of anti-p27kip1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for p27kip1

This gene encodes a cyclin-dependent kinase inhibitor, which shares a limited similarity with CDK inhibitor CDKN1A/p21. The encoded protein binds to and prevents the activation of cyclin E-CDK2 or cyclin D-CDK4 complexes, and thus controls the cell cycle progression at G1. The degradation of this protein, which is triggered by its CDK dependent phosphorylation and subsequent ubiquitination by SCF complexes, is required for the cellular transition from quiescence to the proliferative state. Mutations in this gene are associated with multiple endocrine neoplasia type IV (MEN4). [provided by RefSeq, Apr 2014]

UniProt Comments for p27kip1

p27Kip1: a cell-cycle regulatory protein that Interacts with cyclin-CDK2 and -CDK4, inhibiting cell cycle progression at G1. May mediate TGF beta-induced g1 arrest. Its degradation is triggered by its CDK dependent phosphorylation and subsequent ubiquitination by SCF complexes, is required for the cellular transition from quiescence to the proliferative state.
