Product Name
HphI, Restriction Enzyme
Full Product Name
HphI
Purity/Purification
Molecular Biology Grade
Preparation and Storage
-20 degree C
Dilution Buffer
10mM Tris-HCl (pH 7.4 at 25 degree C), 100mM KCl, 1mM EDTA, 1mM DTT, 0.2mg/ml BSA and 50% glycerol, or Storage Buffer.
Methylation Effects
HphI does not cut GGTGm6A. Blocked by overlapping Dam and EcoBI methylation.

17
pACYC184
16
Labeled Oligonucleotide (LO) Assay
No detectable degradation of a single-stranded and double-stranded labeled oligonucleotide was observed after incubation with 10 units of restriction endonuclease for 4 hours.
Lambda
168
Ligation/Recutting Assay
After 50-fold overdigestion (3u/ug DNA x 17 hours) with HphI, more than 70% of the DNA fragments can be ligated at a 5'-termini concentration of 1.7uM. More than 90% of these can be recut.

Overdigestion Assay
No detectable change in the specific fragmentation pattern is observed after 80-fold overdigestion (5u/ug lambda DNA x 16 hours) with HphI (see Star Activity).

Stability During Prolonged Incubation
A minimum of 0.1 units of enzyme is required for complete digestion of 1ug of lambda DNA in 16 hours at 37 degree C.
Storage Buffer
10mM Tris-HCl (pH 7.5 at 25 degree C), 50mM KCl, 1mM DTT, 0.1mM EDTA, 0.2mg/ml BSA and 50% glycerol.
Thermal Inactivation
Enzyme is inactivated by incubation at 65 degree C for 20min.
Unit Definition
One unit is defined as the amount of enzyme required to digest 1ug of lambda DNA dam-in 1 hour at 37 degree C in 50ul of assay buffer.

Testing Data of HphI restriction enzyme

Restriction Enzyme – Cat# MBS638366

Other Notes

Small volumes of HphI restriction enzyme vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
