Polyglutamylation Modification Antibody – Mouse Monoclonal

For Research Use Only. Not for use in diagnostic procedures. Catalog #: MBS642422

Polyglutamylation is a post-translational modification in which glutamate side chains of variable lengths are formed on the modified protein. It is evolutionarily conserved from and the most prominent substrate is tubulin, the microtubule (MT) building block.… More details

Specificity
Recognizes the posttranslational modification (poly)glutamylation on proteins. Reacts with polyglutamylated alpha- and beta-tubulin.
Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
Octapeptide EGEGE*EEG, modified by the addition of two glutamyl units onto the fifth E (indicated by an asterisk).
Applications Tested/Suitable for anti-Polyglutamylation Modification antibody
Western Blot (WB), Immunoprecipitation (IP), Immunocytochemistry (ICC)
Suitable for use in Immunocytochemistry, Immunoprecipitation, Western Blot

Immunofluorescence (IF) of anti-Polyglutamylation Modification antibody

Immunofluorescence analysis of tubulin glutamylation with MBS642422. HeLa cells grown in standard culture conditions are transfected with GFP-fusion expression plasmids for the polyglutamylases TTLL4 and TTLL6. Glutamylated microtubules are detected with MBS642422. Only in the cells expressing glutamylases (red arrowheads), strong microtubule labelling is detected, whereas in non-transfected cells (white arrowheads), only very weak labelling is seen. Note that the level of endogenous microtubule glutamylation can vary between different cell lines. Picture courtesy of Dr. Carsten Janke, Curie Institute, Paris.

Western Blot (WB) of anti-Polyglutamylation Modification antibody

Western blot analysis of protein glutamylation with MBS642422 to polyglutamylation modification .|Method: HeLa cells grown in standard culture conditions are lysed in SDS sample buffer and run on 10% SDS-PAGE. The proteins are transferred to nitrocellulose membrane and detected by standard Western blot protocol, using the MAb to polyglutamylation modification (1:1’000) in TBS containing 0.1% Tween 20 for washing steps and 5% fat free milk for antibody incubation.is

Testing Data of anti-Polyglutamylation Modification antibody

In HeLa cells, only two non-tubulin proteins are detected (Regnard et al., 2000; van Dijk et al., 2008), whereas after the expression of either TTLL4 or TTLL6 polyglutamylases (van Dijk et al., 2007), tubulin and additional proteins are detected. Brain tubulin is highly polyglutamylated and therefore strongly detected with the 045161|Picture courtesy of Dr Carsten Janke, Curie Institute, Par

Other Notes

Small volumes of anti-Polyglutamylation Modification antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
