Ciliary Neurotrophic Factor (CNTF) Active Protein

For Research Use Only. Not for use in diagnostic procedures. Catalog #: MBS650758

A neurotrophic factor that promotes the survival of various neuronal cell types and may play an important role in the injury response in the nervous system. CNTF, like FGF acidic, FGF basic, and PD-ECGF (platelet-derived endothelial cell growth factor), does… More details

Product Overview

Online Datasheet in PDF
Product Name
Ciliary Neurotrophic Factor (CNTF), Active Protein
Full Product Name
Ciliary Neurotrophic Factor, Recombinant, Human (CNTF)
Product Gene Name
Sequence
The sequence of the first five N-terminal amino acids was determined and was found to be Met-Ala-Ph e-Thr-Glu.
Host
E Coli
Form/Format
Supplied as a lyophilized powder in 5mM sodium phosphate buffer, pH 7.5, 5mM sodium chloride.
Purity/Purification
Highly Purified
98% by RP-HPLC, FPLC, or reducing/non-reducing SDS-PAGE Silver Stain. Chromatographically purified.
Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. Reconstitute to nominal volume by adding sterile dH2O and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer.
Molecular Weight
22,931 Da
Chromosome Location
Chromosome: 11; NC_000011.9 (58390146..58393206). Location: 11q12.2
OMIM
118945
3D Structure
ModBase 3D Structure for P26441
Active Protein Note
Protein activity can be an ambiguous term. Please refer to the data sheet or inquire with Technical Support to determine the definition of activity for this product.
Biological Activity
Recombinant Human Ciliary Neurotrophic Factor is fully biologically active when compared to standard. The ED50 as determined by the dose-dependent stimulation of TF-1 cells is less then 2 ng/ml, corresponding to a Specific Activity of 5 x 10 IU/mg.
Dimers and Aggregates
1% as determined by silver-stained SDS-PAGE gel analysis.
Endotoxin Level
0.1ng/ug (IEU/ug)
Protein Content
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280nm using the absorbency value of 1.28 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC, using a standard solution of human CNTF as a Reference Standard.
Reconstitution
Recommended to reconstitute the lyophilized CNTF in sterile 18M -cm H2O not less than 100ug/ml, which can then be further diluted to other aqueous solutions.

Other Notes

Small volumes of CNTF active protein vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for CNTF

The protein encoded by this gene is a polypeptide hormone whose actions appear to be restricted to the nervous system where it promotes neurotransmitter synthesis and neurite outgrowth in certain neuronal populations. The protein is a potent survival factor for neurons and oligodendrocytes and may be relevant in reducing tissue destruction during inflammatory attacks. A mutation in this gene, which results in aberrant splicing, leads to ciliary neurotrophic factor deficiency, but this phenotype is not causally related to neurologic disease. A read-through transcript variant composed of the upstream ZFP91 gene and CNTF sequence has been identified, but it is thought to be non-coding. Read-through transcription of ZFP91 and CNTF has also been observed in mouse. [provided by RefSeq]
