Product Overview

Online Datasheet in PDF
Product Name
Interleukin 13 (IL13), Active Protein
Full Product Name
Interleukin 13, Recombinant, Human (IL-13)
Product Gene Name
Sequence
The sequence of the first five N-terminal amino acids was determined and found to be Ser-Pro-Gl y-Pro-Val.
Host
E Coli
Form/Format
Supplied as a lyophilized powder without additives.
Purity/Purification
Highly Purified
95% as determined by RP-HPLC, anion-exchange FPLC and/or reducing and non-reducing SDS-PAGE Silver Stained gel.
Preparation and Storage
-20 degree C
Stability: Lyophilized Recombinant Human Interleukin-13 although stable at room temperature for 3 weeks, should be stored desiccated below -180C. Upon reconstitution Human Interleukin-13 should be stored at 40C between 2-7 days and for future use below -180C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please avoid freeze-thaw cycles.
Molecular Weight
12.5kD
Chromosome Location
Chromosome: 5; NC_000005.9 (131993865..131996801). Location: 5q31
OMIM
147683
3D Structure
ModBase 3D Structure for P35225
Active Protein Note
Protein activity can be an ambiguous term. Please refer to the data sheet or inquire with Technical Support to determine the definition of activity for this product.
Biological Activity
Human IL-13 is fully biologically active when compared to standard. The ED50 was determined by the dose dependent prolifiration of TF-1 cells and was found to be < 1ng/ml, corresponding to a specific activity of >1 x 106.
Dimers and Aggregates
Less than 1% as determined by silver-stained SDS-PAGE gel analysis.
Endotoxin Level
0.1ng/ug (IEU/ug) of recombinant Human IL-13.
Protein Content
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280 nm using the absorbency value of 0.57 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC, using a calibrated solution of IL-13 as a Reference Standard.
Solubility
It is recommended to reconstitute the lyophilized Human IL-13 in sterile 18M -cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.

Other Notes

Small volumes of IL13 active protein vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for IL13

This gene encodes an immunoregulatory cytokine produced primarily by activated Th2 cells. This cytokine is involved in several stages of B-cell maturation and differentiation. It up-regulates CD23 and MHC class II expression, and promotes IgE isotype switching of B cells. This cytokine down-regulates macrophage activity, thereby inhibits the production of pro-inflammatory cytokines and chemokines. This cytokine is found to be critical to the pathogenesis of allergen-induced asthma but operates through mechanisms independent of IgE and eosinophils. This gene, IL3, IL5, IL4, and CSF2 form a cytokine gene cluster on chromosome 5q, with this gene particularly close to IL4. [provided by RefSeq]

UniProt Comments for IL13

Function: Cytokine. Inhibits inflammatory cytokine production. Synergizes with IL2 in regulating interferon-gamma synthesis. May be critical in regulating inflammatory and immune responses.

Subunit structure: Interacts with IL13RA2.

Subcellular location: Secreted.

Involvement in disease: Defects in IL13 may be a cause of susceptibility to allergic rhinitis (ALRH) [

MIM:607154]. Allergic rhinitis is a common disease of complex inheritance and is characterized by mucosal inflammation caused by allergen exposure.

Sequence similarities: Belongs to the IL-4/IL-13 family.

Caution: It is uncertain whether Met-1 or Met-15 is the initiator.

Sequence caution: The sequence AAA36107.1 differs from that shown. Reason: Erroneous initiation. Translation N-terminally extended. The sequence AAA83738.1 differs from that shown.
