Sequence
The sequence of the first five N-terminal amino acids was determined and was found to be Met-Ala-Pr o-Thr-Arg.
Host
E Coli
Form/Format
Supplied as a lyophilized powder without additives. Reconstitute in sterile ddH20, 0.1% BSA or HSA to 100ug/ml, which can then be further diluted to other aqueous solutions.
Purity/Purification
Highly Purified
95% by RP-HPLC or reducing/non-reducing SDS-PAGE Silver Stain. Chromatographically purified.
Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. Reconstitute to nominal volume by adding sterile ddH20, 0.1% BSA or HSA, aliquot and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Active Protein Note
Protein activity can be an ambiguous term. Please refer to the data sheet or inquire with Technical Support to determine the definition of activity for this product.
Biological Activity
The ED50 as determined by the dose-dependent stimulation of the proliferation of murine MC/9 cells is 0.001-0.01ng/ml.
Protein Content
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280nm using the absorbency value of 0.966 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC, using a calibrated solution of Rat GM-CSF as a Reference Standard.

Other Notes

Small volumes of CSF2 active protein vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for CSF2

The protein encoded by this gene is a cytokine that controls the production, differentiation, and function of granulocytes and macrophages. The active form of the protein is found extracellularly as a homodimer. This gene has been localized to a cluster of related genes at chromosome region 5q31, which is known to be associated with interstitial deletions in the 5q- syndrome and acute myelogenous leukemia. Other genes in the cluster include those encoding interleukins 4, 5, and 13. [provided by RefSeq]

UniProt Comments for CSF2

Function: Cytokine that stimulates the growth and differentiation of hematopoietic precursor cells from various lineages, including granulocytes, macrophages, eosinophils and erythrocytes.

Subunit structure: Monomer.

Subcellular location: Secreted.

Polymorphism: Variant Ile-117 may be a risk factor for atopic asthma.

Pharmaceutical use: Available under the names Leukine (Immunex) and Leucomax (Novartis). Used in myeloid reconstitution following bone marrow transplant, bone marrow transplant engraftment failure or delay, mobilization and following transplantation of autologous peripheral blood progenitor cells, and following induction chemotherapy in older adults with acute myelogenous leukemia.

Sequence similarities: Belongs to the GM-CSF family.
