Keratinocyte Growth Factor 2 (FGF10) Active Protein

For Research Use Only. Not for use in diagnostic procedures. Catalog #: MBS650907

KGF is a potent specific mitogen for many epithelial cells but not for fibroblasts and endothelial cells. KGF is thought to play an important role in the paracrine growth control of normal epithelial cells. Recombinant Human KGF-2 (FGF-10) is a single,… More details

Product Overview

Online Datasheet in PDF
Product Name
Keratinocyte Growth Factor 2 (FGF10), Active Protein
Full Product Name
Keratinocyte Growth Factor 2, Recombinant, Human (KGF2)
Product Gene Name
Host
E Coli
Form/Format
Supplied as a lyophilized powder in 5mM sodium phosphate, pH 7.4, 80mM sodium chloride.
Purity/Purification
Highly Purified
95% by RP-HPLC, FPLC, or reducing/non-reducing SDS-PAGE Silver Stain. Chromatographically purified.
Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. Reconstitute to nominal volume by adding sterile dH2O and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer.

Chromosome: 5; NC_000005.9 (44305097..44388784, complement). Location: 5p13-p12
OMIM
149730
3D Structure
ModBase 3D Structure for O15520
Active Protein Note
Protein activity can be an ambiguous term. Please refer to the data sheet or inquire with Technical Support to determine the definition of activity for this product.
Biological Activity
Recombinant Keratinocye Growth Factor-2 is fully biologically active when compared to standards. The ED50, calculated by the dose-dependent stimulation of FGF recpetors by BaF3 indicator cells (measured by 3H-thymidine uptake) is less then 0.5ng/ml corresponding to a specific activity of 2 x 10e6 Units/mg.
Dimers and Aggregates
1% as determined by silver-stained SDS-PAGE gel analysis.
Endotoxin Level
0.1ng/ug (IEU/ug) of KGF-2.
Protein Content
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280nm using the absorbency value of 1.79 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC, using a calibrated solution of KGF-2 as a Reference Standard.
Reconstitution
Reconstitute the lyophilized KGF-2 in sterile 18M -cm H2O not less than 100ug/ml, which can then be further diluted to other aqueous solutions.

Other Notes

Small volumes of FGF10 active protein vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for FGF10

The protein encoded by this gene is a member of the fibroblast growth factor (FGF) family. FGF family members possess broad mitogenic and cell survival activities, and are involved in a variety of biological processes, including embryonic development, cell growth, morphogenesis, tissue repair, tumor growth and invasion. This protein exhibits mitogenic activity for keratinizing epidermal cells, but essentially no activity for fibroblasts, which is similar to the biological activity of FGF7. Studies of the mouse homolog of suggested that this gene is required for embryonic epidermal morphogenesis including brain development, lung morphogenesis, and initiation of lim bud formation. This gene is also implicated to be a primary factor in the process of wound healing. [provided by RefSeq]

UniProt Comments for FGF10

Function: Could be a growth factor active in the process of wound healing. Acts as a mitogen in the lung. May act in a manner similar to FGF-7.

Subunit structure: Interacts with FGFBP1. Ref.7

Subcellular location: Secreted

Potential.

Involvement in disease: Defects in FGF10 are the cause of autosomal dominant aplasia of lacrimal and salivary glands (ALSG) [

MIM:180920]. ALSG has variable expressivity, and affected individuals may have aplasia or hypoplasia of the lacrimal, parotid, submandibular and sublingual glands and absence of the lacrimal puncta. The disorder is characterized by irritable eyes, recurrent eye infections, epiphora (constant tearing) and xerostomia (dryness of the mouth), which increases the risk of dental erosion, dental caries, periodontal disease and oral infections. Ref.9Defects in FGF10 are a cause of lacrimo-auriculo-dento-digital syndrome (LADDS) [

MIM:149730]; also known as Levy-Hollister syndrome. LADDS is a form of ectodermal dysplasia, a heterogeneous group of disorders due to abnormal development of two or more ectodermal structures. LADDS is an autosomal dominant syndrome characterized by aplastic/hypoplastic lacrimal and salivary glands and ducts, cup-shaped ears, hearing loss, hypodontia and enamel hypoplasia, and distal limb segments anomalies. In addition to these cardinal features, facial dysmorphism, malformations of the kidney and respiratory system and abnormal genitalia have been reported. Craniosynostosis and severe syndactyly are not observed. Ref.10 Ref.11

Sequence similarities: Belongs to the heparin-binding growth factors family.
