Performance
Five x 105 cultured HPB-MLT cells were washed and incubated 45 minutes on ice with 80 ul of anti-CD28/R-PE at a 1:50 dilution (20ug/ml). Cells were washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 1.88 log10 fluorescent units when compared to a Mouse IgG1/R-PE negative control at a similar concentration. Binding was blocked when cells were pre incubated 10 minutes with 20 ul of 0.5 mg/ml anti-CD28 antibody.
Conjugation
PE
Production
Protein A purified antibody from tissue culture supernatant was conjugated to R-Phycoerythrin through a sulfo-ester linkage. Unconjugated antibody was removed using size exclusion chromatography. Antibody conjugate is at 1.0 mg/ml with an A565/A280 ratio of 3.03.

UniProt Comments for CD28

CD28: a type I plasma membrane protein Involved in T-cell activation, the induction of cell proliferation and cytokine production and promotion of T-cell survival. Binds to B7-1 and -2. Expressed in T cells and plasma cells, but not in less mature B cells. Six splice-variant isoforms have been described.

Protein type: Cell surface; Membrane protein, integral

Cellular Component: membrane; plasma membrane; integral to membrane; immunological synapse; lipid raft; external side of plasma membrane

Molecular Function: protein homodimerization activity; SH3/SH2 adaptor activity; protease binding

Biological Process: positive regulation of isotype switching to IgG isotypes; T cell activation; positive regulation of mitosis; positive regulation of interleukin-2 biosynthetic process; negative thymic T cell selection; T cell receptor signaling pathway; positive regulation of phosphoinositide 3-kinase cascade; positive regulation of inflammatory response to antigenic stimulus; positive regulation of protein kinase B signaling cascade; T cell costimulation; positive regulation of alpha-beta T cell proliferation; positive regulation of T cell proliferation; immune response; positive regulation of transcription from RNA polymerase II promoter; regulatory T cell differentiation
