CD14 Antibody – Mouse Monoclonal

For Research Use Only. Not for use in diagnostic procedures. Catalog #: MBS666359

Information: Human CD14 is strongly expressed on myelomonocyte lineage cells including monocytes, macrophages, and a subset of granulocytes. CD14 is a high affinity receptor for complexes of lipopolysaccharide and lipopolysaccharide binding protein. A soluble… More details

Volume: 200 ul (lot specific)
Preparation and Storage
Store at 2 - 5 degree C. Freeze/Thawing is not recommended. Protect from light
Product should retain activity for at least 12 months after shipping date when stored as recommended.
Molecular Weight
39,667 Da
Performance
Five x 105 ficoll prepared human peripheral blood mononuclear cells were washed and pre incubated 5 minutes with 20mul of 250mug/ml human IgG (to block non specific binding) after which they were incubated 45 minutes on ice with 80 mul of anti-CD14/FITC at a 1:50 dilution (10 mug/ml). Cells were washed three times, fixed and analyzed by FACS. A net 9.7% sub population of the cells stained positive with a mean shift of 1.82 log10 fluorescent units when compared to a Mouse IgG2a/FITC negative control at a similar concentration. Binding was blocked when cells were pre incubated 10 minutes with 20 mul of 0.5 mg/ml anti-CD14 antibody.
Conjugation
FITC
Production
Antibody from (low FBS containing) tissue culture supernatant was Protein A purified to 95% Immunoglobulin by SDS-PAGE, and reacted with FITC. Unconjugated FITC was removed from conjugate using a desalting column. Antibody conjugate is at approximately 0.5 mg/ml with a Fluorescein/Antibody molar ratio of 15.0
Buffer
50 mM Sodium Phosphate pH 7.5, 100 mM Potassium Chloride, 150mM NaCl, 5% Glycerol, 0.2% BSA, 0.04% NaN3 (as a preservative).

Human thymocytes/Sezary T cells
Applications Tested/Suitable for anti-CD14 antibody
Flow Cytometry (FC/FACS)
Application Notes for anti-CD14 antibody
1:50 (or use 1.6mul of concentrated stock per 5 x 105-cell test)

UniProt Comments for CD14

Coreceptor for bacterial lipopolysaccharide. In concert with LBP, binds to monomeric lipopolysaccharide and delivers it to the LY96/TLR4 complex, thereby mediating the innate immune response to bacterial lipopolysaccharide (LPS). Acts via MyD88, TIRAP and TRAF6, leading to NF-kappa-B activation, cytokine secretion and the inflammatory response. Acts as a coreceptor for TLR2:TLR6 heterodimer in response to diacylated lipopeptides and for TLR2:TLR1 heterodimer in response to triacylated lipopeptides, these clusters trigger signaling from the cell surface and subsequently are targeted to the Golgi in a lipid-raft dependent pathway. Binds electronegative LDL (LDL-) and mediates the cytokine release induced by LDL- ().
