Keratin 191
Keratins are a family of intermediate filament proteins that assemble into filaments through forming heterodimers of one type I keratin (keratins 9 to 23) and one type II keratin (keratins 1-8). The two keratin types share only 30% sequence homology.2-6 Keratins demonstrate tissue- and differentiation-specific expression profiles. Cytokeratin 19 is the smallest human keratin, is found in many simple and some non-keratinizing stratified squamous epithelia, and is often co-expressed with keratin 7.7,8 Cytokeratin 19 has been implicated as a marker for stem cells in some tissues, such as hair follicles in the skin.
Monoclonal antibodies to cytokeratin proteins can be useful markers for tumor identification and classification. The degree of cytokeratin 19 positivity in breast cancer distinguishes malignant from benign tumors. Cytokeratin 19 is also associated with malignant transformation in esophageal and pancreatic cancers.9,10
Preparation and Storage
The purified (UNLB) antibody is supplied as 0.1 mg purified immunoglobulin in 1.0 mL of 100 mM borate buffered saline, pH 8.2; store at 2-8 degree C.
The fluorescein (FITC) conjugate is supplied as 0.1 mg in 0.2 mL PBS/NaN3. Store at 2-8 degree C.
The horseradish peroxidase (HRP) conjugate is supplied as 1.0 mL of stock solution in 50% glycerol/50% PBS, pH 7.4. No preservative added. Store at 2-8 degree C or long-term at -20.
Immunogen
Recombinant C-terminal keratin 19
Characterization
To ensure acceptable performance, each batch of product is tested ELISA, immunohistochemical staining (HT29 and HCC38 cells) or Western blotting (HT29 and HCC38 cell lysates) to conform to characteristics of a standard reference reagent.

Other Notes

Small volumes of anti-Cytokeratin 19 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

Protein Family

UniProt Comments for Cytokeratin 19

K19: a type I cytoskeletal keratin. The keratins are intermediate filament proteins responsible for the structural integrity of epithelial cells and are subdivided into cytokeratins and hair keratins. Keratin 19 differs from all other IF proteins in lacking the C-terminal tail domain. Unlike its related family members, this smallest known acidic cytokeratin is not paired with a basic cytokeratin in epithelial cells. It is specifically expressed in the periderm, the transiently superficial layer that envelopes the developing epidermis.

Protein type: Cytoskeletal

Chromosomal Location of Human Ortholog: 17q21.2

Cellular Component: dystrophin-associated glycoprotein complex; costamere; intermediate filament; Z disc; sarcolemma

Molecular Function: protein binding; structural constituent of cytoskeleton; structural constituent of muscle; protein complex binding

Biological Process: viral reproduction; response to estrogen stimulus; sarcomere organization
