Application Notes for anti-IL17A antibody
Research Application(s): ELISA Capture: TC11-18H10 monoclonal antibody is useful as a capture antibody in a sandwich ELISA for quantifying mouse IL-17 protein levels. Purified TC11-18H10 antibody should be paired with biotinylated TC11-8H4 antibody as the detection antibody, with purified recombinant mouse IL-17 as the standard.
Immunofluorescence/Flow Cytometry: TC11-18H10 antibody is useful for intracytoplasmic staining and flow cytometric analysis to identify and enumerate IL-17-positive cells within mixed cell populations. Rat IgG1 should be used as a specificity control. Western Blotting: The purified TC11-18H10 antibody is also useful for Western blotting applications. For Western blotting, a concentration of 1-5 ug/mL (in conjunction with AP-labeled Goat Anti-Rat-Ig) has been found to enable detection of <= 100 ng/lane of recombinant mouse IL-17 under reducing conditions.
Neutralization: The LE/AF form of TC11-18H10 can be used for neutralizing mouse IL-17 bioactivity. A suitable LE/AF Rat IgG1 should be used as an isotype control.
Since applications vary, each investigator should determine the optimal concentration appropriate for individual applications.

Other Notes

Small volumes of anti-IL17A antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for IL17A

The protein encoded by this gene is a proinflammatory cytokine produced by activated T cells. This cytokine regulates the activities of NF-kappaB and mitogen-activated protein kinases. This cytokine can stimulate the expression of IL6 and cyclooxygenase-2 (PTGS2/COX-2), as well as enhance the production of nitric oxide (NO). High levels of this cytokine are associated with several chronic inflammatory diseases including rheumatoid arthritis, psoriasis and multiple sclerosis. [provided by RefSeq, Jul 2008]
