Application Notes for anti-IFNG antibody
Research Application(s): ELISA Detection: XMG1.2 monoclonal antibody is useful as a capture antibody in a sandwich ELISA for quantifying mouse IFN- gamma protein levels. Biotinylated XMG1.2 antibody should be paired with a suitable purified antibody as the capture antibody, with purified recombinant human IFN- gamma as the standard.
Western Blotting: The purified XMG1.2 antibody is also useful for Western blotting applications. For Western blotting, a concentration of 1-5 ug/mL (in conjunction with AP-labeled Goat Anti-Rat-Ig) has been found to enable detection of <= 100 ng/lane of recombinant mouse IFN- gamma under reducing conditions.
Immunofluorescence/Flow Cytometry: XMG1.2 antibody is useful for intracytoplasmic staining and flow cytometric analysis to identify and enumerate IFN- gamma -positive cells within mixed cell populations.4-6 Recombinant mouse IFN- gamma or Rat IgG1 should be used as a specificity control. FITC conjugated XMG1.2 antibodies are useful for intracellular staining in cells producing IFN- gamma. our intracellular staining buffers are recommended for this application.
Neutralization: The LE/AF form of XMG1.2 can be used for neutralizing mouse IFN- gamma bioactivity. A suitable LE/AF Rat IgG1 should be used as an isotype control.
Since applications vary, each investigator should determine the optimal concentration appropriate for individual applications.

Other Notes

Small volumes of anti-IFNG antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for IFNG

This gene encodes a member of the type II interferon family. The protein encoded is a soluble cytokine with antiviral, immunoregulatory and anti-tumor properties and is a potent activator of macrophages. Mutations in this gene are associated with aplastic anemia.

UniProt Comments for IFNG

Function: Produced by lymphocytes activated by specific antigens or mitogens. IFN-gamma, in addition to having antiviral activity, has important immunoregulatory functions. It is a potent activator of macrophages, it has antiproliferative effects on transformed cells and it can potentiate the antiviral and antitumor effects of the type I interferons.

Subunit structure: Homodimer.

Subcellular location: Secreted.

Tissue specificity: Released primarily from activated T lymphocytes.

Post-translational modification: Proteolytic processing produces C-terminal heterogeneity, with proteins ending alternatively at Gly-150, Met-157 or Gly-161.

Involvement in disease: In Caucasians, genetic variation in IFNG is associated with the risk of aplastic anemia (AA) [

MIM:609135]. AA is a rare disease in which the reduction of the circulating blood cells results from damage to the stem cell pool in bone marrow. In most patients, the stem cell lesion is caused by an autoimmune attack. T-lymphocytes, activated by an endogenous or exogenous, and most often unknown antigenic stimulus, secrete cytokines, including IFN-gamma, which would in turn be able to suppress hematopoiesis.

Pharmaceutical use: Available under the name Actimmune (Genentech). Used for reducing the frequency and severity of serious infections associated with chronic granulomatous disease (CGD).

Sequence similarities: Belongs to the type II (or gamma) interferon family.
