CD19 is a monomeric transmembrane glycoprotein expressed at relatively constant levels throughout B cell development from early pro-B/pre-B cells (i.e. B220^+/CD43^+/HSA^+) through fully differentiated B cell stages. ^1-7 Terminally differentiated plasma cells do not express CD19. ^1 In humans, the CD19 molecule on the surface of mature B cells associates with CD21 (CR-2) and CD81 (TAPA-1), and this multimolecular complex synergizes with surface immunoglobulin to provide signal transduction and promote cellular activation. ^3, 4 All splenic and peritoneal IgM^+ cells of both B-1 and B-2 lineages are CD19^+, with B-1 cells expressing higher levels of CD19 than B-2 cells in these sites.1, 7 Recent studies with CD19-deficient mice have suggested that this molecule may not be required for normal generation and maturation of B cells in the bone marrow. ^7 Monoclonal antibody 6D5 recognizes the same epitope as the published clone, 1D3. ^1, 8

NCBI Summary for CD19

Lymphocytes proliferate and differentiate in response to various concentrations of different antigens. The ability of the B cell to respond in a specific, yet sensitive manner to the various antigens is achieved with the use of low-affinity antigen receptors. This gene encodes a cell surface molecule which assembles with the antigen receptor of B lymphocytes in order to decrease the threshold for antigen receptor-dependent stimulation. UniProt Comments for CD19

Function: Assembles with the antigen receptor of B lymphocytes in order to decrease the threshold for antigen receptor-dependent stimulation.

Subunit structure: Forms a complex with CD21, CD81 and CD225 in the membrane of mature B cells. Interacts with VAV. Interacts with GRB2 and SOS when phosphorylated on Tyr-348 and/or Tyr-378. Interacts with PLCG2 when phosphorylated on Tyr-409. Ref.11

Subcellular location: Membrane; Single-pass type I membrane protein.

Post-translational modification: Phosphorylated on serine and threonine upon DNA damage, probably by ATM or ATR. Phosphorylated on tyrosine following B-cell activation. Ref.10 Ref.11 Ref.12

Involvement in disease: Defects in CD19 are the cause of immunodeficiency common variable type 3 (CVID3) [

MIM:613493]; also called antibody deficiency due to CD19 defect. CVID3 is a primary immunodeficiency characterized by antibody deficiency, hypogammaglobulinemia, recurrent bacterial infections and an inability to mount an antibody response to antigen. The defect results from a failure of B-cell differentiation and impaired secretion of immunoglobulins; the numbers of circulating B cells is usually in the normal range, but can be low. Ref.13

Sequence similarities: Contains 2 Ig-like C2-type (immunoglobulin-like) domains.

Sequence caution: The sequence AAA35533.1 differs from that shown. Reason: Frameshift at position 396.
