Application Notes for anti-MCP-1 antibody
Immunohistochemistery: This antibody stained formalin-fixed, paraffin-embedded sections of human breast invasive ductal carcinoma. The recommended concentration is 10.0 mug/ml with an overnight incubation at 4 degree C. An HRP-labeled polymer detection system was used with a DAB chromogen. Heat induced antigen retrieval with a pH 6.0 sodium citrate buffer is recommended. Tissue samples were provided by the Cooperative Human Tissue Network, which is funded by the National Cancer Institute.

Sandwich ELISA: In a sandwich ELISA (assuming 100 ul/well), a concentration of 2.0-4.0 ug/ml of this antibody will detect at least 300 pg/ml of recombinant human MCP-1 when used with biotinylated antigen affinity purified anti-human MCP-1 as the detection antibody at a concentration of approximately 0.5-1.0 ug/ml.

Western Blot: To detect hMCP-1 by Western Blot analysis this antibody can be used at a concentration of 0.20-0.40 ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hMCP-1 is 1.0-2.0 ng/lane, under reducing or non-reducing conditions.

Other Notes

Small volumes of anti-MCP-1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for MCP-1

This gene is one of several cytokine genes clustered on the q-arm of chromosome 17. Chemokines are a superfamily of secreted proteins involved in immunoregulatory and inflammatory processes. The superfamily is divided into four subfamilies based on the arrangement of N-terminal cysteine residues of the mature peptide. This chemokine is a member of the CC subfamily which is characterized by two adjacent cysteine residues. This cytokine displays chemotactic activity for monocytes and basophils but not for neutrophils or eosinophils. It has been implicated in the pathogenesis of diseases characterized by monocytic infiltrates, like psoriasis, rheumatoid arthritis and atherosclerosis. It binds to chemokine receptors CCR2 and CCR4. UniProt Comments for MCP-1

CCL2: Chemotactic factor that attracts monocytes and basophils but not neutrophils or eosinophils. Augments monocyte anti-tumor activity. Has been implicated in the pathogenesis of diseases characterized by monocytic infiltrates, like psoriasis, rheumatoid arthritis or atherosclerosis. May be involved in the recruitment of monocytes into the arterial wall during the disease process of atherosclerosis. Monomer or homodimer; in equilibrium. Binds to CCR2 and CCR4. Is tethered on endothelial cells by glycosaminoglycan (GAG) side chains of proteoglycans. Belongs to the intercrine beta (chemokine CC) family.
