Application Notes for anti-NANOG antibody
Immunohistochemistery: This antibody stained formalin-fixed, paraffin-embedded sections of human prostate malignant adenocarcinoma. The recommended concentration is 0.5 mg/ml with an overnight incubation at 4 degree C. An alkaline phosphatase-labeled polymer detection system was used with a non-alcohol soluble red chromogen. Heat induced antigen retrieval with a pH 6.0 sodium citrate buffer is recommended.

Sandwich ELISA: To detect hNanog by sandwich ELISA (using 100 mul/well antibody solution) a concentration of 0.5 - 2.0 mug/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with Biotinylated Anti-Human Nanog as a detection antibody, allows the detection of at least 0.2 - 0.4 ng/well of recombinant hNanog.

Western Blot: To detect hNanog by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hNanog is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.

Other Notes

Small volumes of anti-NANOG antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for NANOG

The protein encoded by this gene is a DNA binding homeobox transcription factor involved in embryonic stem (ES) cell proliferation, renewal, and pluripotency. The encoded protein can block ES cell differentiation and can also autorepress its own expression in differentiating cells. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Sep 2015]

UniProt Comments for NANOG

NANOG: Transcription regulator involved in inner cell mass and embryonic stem (ES) cells proliferation and self-renewal. Imposes pluripotency on ES cells and prevents their differentiation towards extraembryonic endoderm and trophectoderm lineages. Blocks bone morphogenetic protein-induced mesoderm differentiation of ES cells by physically interacting with SMAD1 and interfering with the recruitment of coactivators to the active SMAD transcriptional complexes. Acts as a transcriptional activator or repressor. Binds optimally to the DNA consensus sequence 5'-TAAT[GT][GT]-3' or 5'-[CG][GA][CG]C[GC]ATTAN[GC]-3'. When overexpressed, promotes cells to enter into S phase and proliferation. Interacts with SMAD1 and SALL4. Expressed in testicular carcinoma and derived germ cell tumors. Expressed in fetal gonads, ovary and testis. Also expressed in ovary teratocarcinoma cell line and testicular embryonic carcinoma. Not expressed in many somatic organs and oocytes. Belongs to the Nanog homeobox family. 2 isoforms of the human protein are produced by alternative splicing.
