Application Notes for anti-IL-8 antibody
Direct Detection: To detect Human IL-8 by direct ELISA (using 100 mul/well antibody solution) a concentration of 0.25 - 1.0 mug/ml of this antibody is required. This biotinylated polyclonal antibody, in conjunction with compatible secondary reagents, allows the detection of at least 0.2 - 0.4 ng/well of recombinant Human IL-8.

Sandwich ELISA: To detect Human IL-8 by sandwich ELISA (using 100 mul/well antibody solution) a concentration of 0.25 - 1.0 mug/ml of this antibody is required. This biotinylated polyclonal antibody, in conjunction with Polyclonal Anti-Human IL-8 as a capture antibody, allows the detection of at least 0.2 - 0.4 ng/well of recombinant Human IL-8.

Western Blot: To detect Human IL-8 by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 ug/ml. When used in conjunction with compatible secondary reagents, the detection limit for recombinant Human IL-8 is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.

Other Notes

Small volumes of anti-IL-8 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for IL-8

The protein encoded by this gene is a member of the CXC chemokine family. This chemokine is one of the major mediators of the inflammatory response. This chemokine is secreted by several cell types. It functions as a chemoattractant, and is also a potent angiogenic factor. This gene is believed to play a role in the pathogenesis of bronchiolitis, a common respiratory tract disease caused by viral infection. This gene and other ten members of the CXC chemokine gene family form a chemokine gene cluster in a region mapped to chromosome 4q. [provided by RefSeq, Jul 2008]

UniProt Comments for IL-8

IL8: IL-8 is a chemotactic factor that attracts neutrophils, basophils, and T-cells, but not monocytes. It is also involved in neutrophil activation. It is released from several cell types in response to an inflammatory stimulus. IL-8(6-77) has a 5-10-fold higher activity on neutrophil activation, IL-8(5-77) has increased activity on neutrophil activation and IL-8(7-77) has a higher affinity to receptors CXCR1 and CXCR2 as compared to IL-8(1-77), respectively. Belongs to the intercrine alpha (chemokine CxC) family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Cytokine; Motility/polarity/chemotaxis; Secreted, signal peptide; Secreted

Chromosomal Location of Human Ortholog: 4q13-q21
