Application Notes for anti-MIP-4 antibody
Immunohistochemistery: This antibody stained formalin-fixed, paraffin-embedded sections of human normal placenta. The recommended concentration is 5.0 ug/mL with an overnight incubation at 4 degree C. An HRP-labeled polymer detection system was used with a DAB chromogen. Heat induced antigen retrieval with a pH 6.0 buffer is recommended. Tissue samples were provided by the Cooperative Human Tissue Network, which is funded by the National Cancer Institute. Information and photo are courtesy of Flagship Biosciences, LLC - Histology Services.

Neutralization: To yield one-half maximal inhibition [ND50] of the biological activity of hMIP-4 (100.0 ng/ml), a concentration of 8.0-10.0 ug/ml of this antibody is required.

Sandwich ELISA: To detect hMIP-4 by sandwich ELISA (using 100 mul/well antibody solution) a concentration of 0.5 - 2.0 mug/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with Biotinylated Anti-Human MIP-4 as a detection antibody, allows the detection of at least 0.2 - 0.4 ng/well of recombinant hMIP-4.

Western Blot: To detect hMIP-4 by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2 ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hMIP-4 is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.

Other Notes

Small volumes of anti-MIP-4 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for MIP-4

This antimicrobial gene is one of several Cys-Cys (CC) cytokine genes clustered on the q arm of chromosome 17. Cytokines are a family of secreted proteins involved in immunoregulatory and inflammatory processes. The CC cytokines are proteins characterized by two adjacent cysteines. The cytokine encoded by this gene displays chemotactic activity for naive T cells, CD4+ and CD8+ T cells and nonactivated lymphocytes, but not for monocytes or granulocytes. This chemokine attracts naive T lymphocytes toward dendritic cells and activated macrophages in lymph nodes. It may play a role in both humoral and cell-mediated immunity responses. UniProt Comments for MIP-4

CCL18: Chemotactic factor that attracts lymphocytes but not monocytes or granulocytes. May be involved in B-cell migration into B-cell follicles in lymph nodes. Attracts naive T-lymphocytes toward dendritic cells and activated macrophages in lymph nodes, has chemotactic activity for naive T-cells, CD4+ and CD8+ T-cells and thus may play a role in both humoral and cell-mediated immunity responses. Belongs to the intercrine beta (chemokine CC) family.

Protein type: Secreted; Secreted, signal peptide; Motility/polarity/chemotaxis; Chemokine

Chromosomal Location of Human Ortholog: 17q12
