Application Notes for anti-FGF antibody
Immunohistochemistery: This antibody stained formalin-fixed, paraffin-embedded sections of human breast invasive ductal carcinoma. The recommended concentrations are 0.25 mug/ml-0.5 mug/ml for two hours at room temperature. An HRP-labeled polymer detection system was used with DAB chromogen. Heat induced antigen retrieval was performed with a pH 6.0 Sodium Citrate buffer.

Neutralization: To yield one-half maximal inhibition [ND50] of the biological activity of Human FGF-basic (0.3 ng/ml), a concentration of 0.25 - 0.40 ug/ml of this antibody is required.

Sandwich ELISA: To detect Human FGF-basic by sandwich ELISA (using 100 mul/well antibody solution) a concentration of 0.5 - 2.0 mug/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with Biotinylated Anti-Human FGF-basic as a detection antibody, allows the detection of at least 0.2 - 0.4 ng/well of recombinant Human FGF-basic.

Western Blot: To detect Human FGF-basic by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant Human FGF-basic is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.

Other Notes

Small volumes of anti-FGF antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for FGF

The protein encoded by this gene is a member of the fibroblast growth factor (FGF) family. FGF family members bind heparin and possess broad mitogenic and angiogenic activities. This protein has been implicated in diverse biological processes, such as limb and nervous system development, wound healing, and tumor growth. The mRNA for this gene contains multiple polyadenylation sites, and is alternatively translated from non-AUG (CUG) and AUG initiation codons, resulting in five different isoforms with distinct properties. The CUG-initiated isoforms are localized in the nucleus and are responsible for the intracrine effect, whereas, the AUG-initiated form is mostly cytosolic and is responsible for the paracrine and autocrine effects of this FGF. UniProt Comments for FGF

FGF2: Plays an important role in the regulation of cell survival, cell division, angiogenesis, cell differentiation and cell migration. Functions as potent mitogen in vitro. Monomer. Homodimer. Interacts with FGFR1, FGFR2, FGFR3 and FGFR4. Affinity between fibroblast growth factors (FGFs) and their receptors is increased by heparan sulfate glycosaminoglycans that function as coreceptors. Interacts with CSPG4, FGFBP1 and TEC. Found in a complex with FGFBP1, FGF1 and FGF2. Expressed in granulosa and cumulus cells. Expressed in hepatocellular carcinoma cells, but not in non- cancerous liver tissue. Belongs to the heparin-binding growth factors family. 4 isoforms of the human protein are produced by alternative initiation.
