Application Notes for anti-MIP-1 alpha antibody
Immunohistochemistery: This antibody stained formalin-fixed, paraffin-embedded sections of human breast malignant carcinoma. The recommended concentration is 5.0 ug/mL with an overnight incubation at 4 degree C. An HRP-labeled polymer detection system was used with a DAB chromogen. Heat induced antigen retrieval with a pH 9.0 buffer is recommended. Tissue samples were provided by the Cooperative Human Tissue Network, which is funded by the National Cancer Institute. Information and photo are courtesy of Flagship Biosciences, LLC - Histology Services.

Sandwich ELISA: To detect Human MIP-1 alpha by sandwich ELISA (using 100 mul/well antibody solution) a concentration of 0.5 - 2.0 mug/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with Biotinylated Anti-Human MIP-1 alpha as a detection antibody, allows the detection of at least 0.2 - 0.4 ng/well of recombinant Human MIP-1 alpha.

Western Blot: To detect Human MIP-1 alpha by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2 ug/ml. When used in conjunction with compatible secondary reagents, the detection limit for recombinant Human MIP-1 alpha is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.

Other Notes

Small volumes of anti-MIP-1 alpha antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for MIP-1 alpha

This locus represents a small inducible cytokine. The encoded protein, also known as macrophage inflammatory protein 1 alpha, plays a role in inflammatory responses through binding to the receptors CCR1, CCR4 and CCR5. Polymorphisms at this locus may be associated with both resistance and susceptibility to infection by human immunodeficiency virus type 1.[provided by RefSeq, Sep 2010]

UniProt Comments for MIP-1 alpha

CCL3: Monokine with inflammatory and chemokinetic properties. Binds to CCR1, CCR4 and CCR5. One of the major HIV-suppressive factors produced by CD8+ T-cells. Recombinant MIP-1-alpha induces a dose-dependent inhibition of different strains of HIV-1, HIV-2, and simian immunodeficiency virus (SIV). Belongs to the intercrine beta (chemokine CC) family.
