Application Notes for anti-IGF-I antibody
Immunohistochemistery: This antibody stained formalin-fixed, paraffin-embedded sections of human breast invasive ductal carcinoma. The recommended concentration is 0.25 ug/ml with an overnight incubation at 4 degree C. An HRP-labeled polymer detection system was used with a DAB chromogen. Optimal results for these conditions were achieved without antigen retrieval.

Neutralization: To yield one-half maximal inhibition [ND50] of the biological activity of hIGF-I (5.0 ng/ml), a concentration of 0.67 - 1.0 ug/ml of this antibody is required.

Sandwich ELISA: To detect hIGF-I by sandwich ELISA (using 100 mul/well antibody solution) a concentration of 0.5 - 2.0 mug/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with Biotinylated Anti-Human IGF-I as a detection antibody, allows the detection of at least 0.2 - 0.4 ng/well of recombinant hIGF-I.

Western Blot: To detect hIGF-I by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hIGF-I is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.

Other Notes

Small volumes of anti-IGF-I antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for IGF-I

The protein encoded by this gene is similar to insulin in function and structure and is a member of a family of proteins involved in mediating growth and development. The encoded protein is processed from a precursor, bound by a specific receptor, and secreted. Defects in this gene are a cause of insulin-like growth factor I deficiency. Several transcript variants encoding different isoforms have been found for this gene.[provided by RefSeq, Mar 2009]

UniProt Comments for IGF-I

IGF1: The insulin-like growth factors, isolated from plasma, are structurally and functionally related to insulin but have a much higher growth-promoting activity. May be a physiological regulator of [1-14C]-2-deoxy-D-glucose (2DG) transport and glycogen synthesis in osteoblasts. Stimulates glucose transport in rat bone-derived osteoblastic (PyMS) cells and is effective at much lower concentrations than insulin, not only regarding glycogen and DNA synthesis but also with regard to enhancing glucose uptake. Defects in IGF1 are the cause of insulin-like growth factor I deficiency (IGF1 deficiency). IGF1 deficiency is an autosomal recessive disorder characterized by growth retardation, sensorineural deafness and mental retardation. Belongs to the insulin family. 3 isoforms of the human protein are produced by alternative splicing.
