Application Notes for anti-sFas antibody
Immunohistochemistery: This antibody stained formalin-fixed, paraffin-embedded sections of human angiosarcoma. The recommended concentration is 0.75 mug/mL - 2.5 mug/mL with an overnight incubation at 4 degree C. An HRP-labeled polymer detection system was used with a AB chromogen. Heat induced antigen retrieval with a pH 6.0 sodium citrate buffer is recommended.

Neutralization: To yield one-half maximal inhibition [ND50] of the biological activity of sFasL/Apo1L (10 ng/ml), a concentration of 0.08 - 0.1 ug/ml of this antibody is required.

Sandwich ELISA: To detect hsFasL/Apo1L by sandwich ELISA (using 100 mul/well antibody solution) a concentration of 0.5 - 2.0 mug/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with Biotinylated Anti-Human sFasL/Apo1L as a detection antibody, allows the detection of at least 0.2 - 0.4 ng/well of recombinant hsFasL/Apo1L.

Western Blot: To detect hsFasL/Apo1L by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hsFasL/Apo1L is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.

Other Notes

Small volumes of anti-sFas antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for sFas

This gene is a member of the tumor necrosis factor superfamily. The primary function of the encoded transmembrane protein is the induction of apoptosis triggered by binding to FAS. The FAS/FASLG signaling pathway is essential for immune system regulation, including activation-induced cell death (AICD) of T cells and cytotoxic T lymphocyte induced cell death. It has also been implicated in the progression of several cancers. Defects in this gene may be related to some cases of systemic lupus erythematosus (SLE). Alternatively spliced transcript variants have been described. [provided by RefSeq, Nov 2014]

UniProt Comments for sFas

FasL: Cytokine that binds to TNFRSF6/FAS, a receptor that transduces the apoptotic signal into cells. May be involved in cytotoxic T-cell mediated apoptosis and in T-cell development. TNFRSF6/FAS-mediated apoptosis may have a role in the induction of peripheral tolerance, in the antigen-stimulated suicide of mature T-cells, or both. Binding to the decoy receptor TNFRSF6B/DcR3 modulates its effects. Homotrimer (Probable). Interacts with ARHGAP9, BAIAP2L1, BTK, CACNB3, CACNB4, CRK, DLG2, DNMBP, DOCK4, EPS8L3, FGR, FYB, FYN, HCK, ITK, ITSN2, KALRN, LYN, MACC1, MIA, MPP4, MYO15A, NCF1, NCK1, NCK2, NCKIPSD, OSTF1, PIK3R1, PSTPIP1, RIMBP3C, SAMSN1, SH3GL3, SH3PXD2B, SH3PXD2A, SH3RF2, SKAP2, SNX33, SNX9, SORBS3, SPTA1, SRC, SRGAP1, SRGAP2, SRGAP3, TEC, TJP3 and YES1. Belongs to the tumor necrosis factor family. 2 isoforms of the human protein are produced by alternative splicing.
